摘 要:目的:观察气滞血瘀证大鼠一氧化氮/一氧化氮合成酶(NO/NOS)的体系变化。方法:将Wistar大鼠随机分成4组,对照组、模型组、血府逐瘀汤低、高剂量组。采用多因素联合刺激法建立气滞血瘀模型。运用分光光度法测定血浆和肝组织中NO含量以及肝组织中NOS活性,利用RT-PCR法测定肝组织中NOS基因表达。结果:模型组血浆NO含量(0.52±0.33)μmol/L显著低于对照组(2.77±1.73)μmol/L(P<0.01),低剂量组NO含量(2.37±0.53)μmol/L显著高于模型组(0.52±0.33)μmol/L(P<0.01);模型组肝组织NO含量(0.52±0.24)μmol/mg显著低于对照组(5.87±1.72)μmol/mg(P<0.01),低剂量组NO含量(3.72±1.53)μmol/mg显著高于模型组(0.52±0.24)μmol/mg(P<0.01);模型组肝组织eNOS基因表达水平显著低于对照组(P<0.01),低剂量组肝组织eNOS显著高于模型组(P<0.05)。结论:气滞血瘀证大鼠NO/NOS体系下调,为进一步研究治疗血瘀症类疾病提供了借鉴和思路。Objective: To investigate change of nitric oxide (NO)/NO synthase (NOS) in Qi-blood stagnation of rats. Methods : Wistar rats were randomized to four groups: normal control group, model group, low concentration group, and high concentration group. The Qi-blood stagnation model was established by complex pathogens. The content of NO in plasma and liver tissues of rats was measured by spectrophotometric. RT-PCR was used to analyze liver tissues NOS mRNA expression. Results: The plasma NO of model group [ (0.52±0.33)μmol/L] decreased significantly compared with normal control group[ (2.77±1.73)μmol/L] (P 〈0.01 ). The plasma NO of treatment group [ ( 2.37±0.53 )μmol/L ] increased significantly compared with model group [ ( 0.52±0.33 ) μmol/L ] ( P 〈 0.01 ). The NO in liver tissues of model group [ (0.52±0.24 ) μmol/mg ] decreased significantly compared with normal control group [ (5.87 ± 1. 72 ) μmol/mg ] ( P 〈 0.01 ). The NO in liver tissues of treatment group [ ( 3.72 ±1.53 ) μmol/mg ] increased significantly compared with model group[ (0.52±0.24) μmoL/mg] (P 〈0.01 ). The expression of NOS in liver tissues NOS mRNA expression in model group de-creased significantly compared with normal control group (P 〈 0.01 ). The expression of NOS in liver tissues NOS mRNA expression in low concentration increased significantly compared with model group (P 〈0.05 ). Conclusion: Nitric oxide( NO)/NO synthase(NOS) was down-regulated in Qi-blood stagnation rats.
