摘 要:目的检测在白细胞介素-6(IL-6)环境培养下前列腺癌细胞微核糖核酸-21(miR-21)的表达,细胞的增殖能力。分析IL-6和miR-21对前列腺癌细胞的作用及意义。方法RT-PCR检测miR-21的表达情况。CCK-8检测细胞增殖能力。通过westernblot检测PDCD4和Bcl-2的表达情况。结果IL-6诱导miR-21的高表达,miR-21靶向PDCD4、Bcl-2等下游基因,从而促进PC-3细胞增殖。结论IL-6通过调控miR-21高表达有可能在前列腺癌中起重要作用。Objective To detect the different expression ofmicroRNA-21 aming prostate carcinoma cells in exposure to distinct concentration of interleukin-6 andits effcet on cell proliferation. Methods RT-PCR was used to detect the expression ofmicroRNA-21 among prostate carcinoma cells exposed to distinct concentration of interleukin-6. CCK-8 was applied to check the capacity of cell proliferation. Expressions of Bcl-2 and PDCD4 were detected by western blot. Results IL - 6 enhanced high expression ofmiR- 21 and miR-21 regulated the target genes of PDCD4 and Bcl-2, further increased the proliferation of PC3 cells. Conclusion IL-6 promoted miR-21 expression,which might play a critical role in prostate cancer.
