摘 要:目的:比较观察年轻小鼠和自然衰老小鼠的骨质特征及骨髓间充质干细胞(bonemarrowmesenchymal stem cells,BMMSCs)的生物学功能。方法:分别取12月龄和2月龄C57BL/6J小鼠,Micro-CT活体扫描两组小鼠股骨骭骺端,三维图像重建后观察其骨质特征;体外分离、培养两组小鼠的BMMSCs,通过MTT、克隆形成和成骨诱导观察其增殖、自我更新和成骨分化能力;并采用实时定量基因扩增荧光检测系统(Real-time PCR)对成骨诱导14 d后的BMMSCs重要相关基因OCN进行检测。结果:与2月龄小鼠相比,12月龄小鼠出现了骨衰老症状,主要表现为骨密度及骨小梁强度下降;BMMSCs的增殖、自我更新和成骨分化能力以及OCN mRNA表达水平均明显降低。结论:BMMSCs可能在骨衰老中发挥了重要作用。AIM: To compare the architecture of trabecular bone and biological function of bone marrow mesenchymal stem cells (BMMSCs) between young and old mice. METHODS: The three dimensional images of tra- becular bone at distal femoral osteoepiphysis of 12 month and 2 month old C57BL/6J mice were observed by Micro-CT. BMMSCs were isolated and cultured from the mice of the 2 groups. The proliferation, renewal and osteogenic differen- tiation function of BMMSCs were observerd by MT'F, clone formation and osteogenic induction assays. BMMSCs were induced by osteogenic medium for 14 days, and then the osteogenesis-associated gene OCN expression was determined by Real - time PCR. RESULTS : Bone mineral density and bone trabecular strength declination were observed at distal femoral osteoepiphysis in 12-months old mice, The abilities of proliferation, renewal, osteogenic differentiation and OCN mRNA expression level were lower in the BMMSCs of 12-months old mice than in those of 2 - moths old mice. CONCLUSION: BMMSCs may play an important role in bone aging.
