摘 要:目的研究血卟啉单甲醚介导的光、声动力联合杀伤C6细胞的效应并对其机制进行初步分析。方法实验分为对照组、声动力组、单纯激光光照组、光动力组和光动力+声动力组。在光、声动力联合组中,首先采用强度为0.5 W/cm2、频率为1 MHz的超声照射,之后应用不同能量密度(40、80、120、160、200和240 J/cm2)的激光进行照射。应用MTT方法检测细胞死亡率,采用流式细胞仪检测细胞凋亡,荧光分析仪检测细胞内活性氧水平的变化。结果超声剂量固定为0.5 W/cm2,频率1 MHz条件下,当激光能量密度小于200 J/cm2时,声、光动力联合组C6细胞的杀伤率明显高于任何单一模式。当激光能量密度为80 J/cm2时,声、光动力联合组所诱导的C6细胞凋亡率最高。声、光动力联合可明显增加细胞内活性氧的产量。结论光、声动力联合能明显增加C6细胞杀伤率。Objective To study the effects and mechanism to kill C6 cells with hematoporphyrin monomethyl ether (HMME)-mediated sonody- namic therapy combining with photodynamic therapy (SPDT). Methods The samples were divided into the control group, SDT group, light irradiation group, PDT group and SPDT group. In the SPDT group, cell were treated with sonication at an intensity of 0. 5 W/cm2 and a frequency of 1 MHz, and then irradiated with differ- ent light fluences. The post-treatment growth-inhibition rates were determined with MTY assay. The apoptotic rates were examined with flow cytometry. The levels of reactive oxygen species (ROS) were measured with fluorescence microplate reader. Results The growth-inhibition rate of C6 glioma cells in the SPDT group was remarkably higher than that in the PDT group when the light dosage was less than 200 J/cm2. The SPDT group displayed the highest apoptosis rate when the light dosage adopted stood at 80 J/ cm2. More ROS was generated in the SPDT group than in the PDT group.Conclusions SPDT can effectively kill C6 glioma cells through synergistic effects of SDT and PDT. C6 glioma cells die due to apopto- sis or necrosis. Abundant ROS may play an important role in SPDT.
