p53基因蛋白在宫颈上皮内瘤样变的表达及意义的研究

摘　　要：目的探讨p53基因蛋白在宫颈上皮内瘤样变(CIN)的表达及意义。方法随机选取2010年3月～2012年9月在本院治疗的100例患者的手术标本为研究对象,实验组60份宫颈上皮内瘤样变标本采用免疫组化染色检测,依据CIN的级别分成3组,对照组40份标本采用TUNEL法检测各组细胞的凋亡情况,分成阳性组(宫颈癌)和阴性组(正常宫颈鳞状上皮);分析5组间的p53阳性表达率。结果正常宫颈鳞状上皮的p53蛋白表达率为0,而宫颈癌p53蛋白表达率为40%,且CIN分级越高,p53阳性表达率也越高,且+++的概率也越高;宫颈癌的细胞分级越高,p53蛋白表达率也越高,即癌细胞的分化率越差,p53蛋白表达率越高,CINⅠ与CINⅡ、Ⅲ比较,差异有统计学意义(P<0.05)。结论 p53基因蛋白表达与CIN的发生、发展有密切关系,是宫颈癌发生的早期事件。Objective To investigate the expression and significance of p53 gene protein-like change in cervical in traepithelial neoplasia （CIN）. Methods The surgical specimens of 100 eases of patients treated in our hospital from March 2010 to September 2012 were selected as the research objects. 60 samples of experimental group with cervical intraepithelial neoplasia-like change were given casesimmunohistochemical staining, and divided into three groups according to the level of CIN, 40 samples in control group using TUNEL assay apoptosis of cells in each group, divided into positive group （cervical cancer） aggregate negative group （normal cervical squamous epithelium）; p53 positive expression rate of the five groups was analyzed. Results p53 protein expression were 0, 40% in normal cervical squamous epithelium and cervical cancer, and in CIN grade, the higher the grading was, the higher the positive expression rate became, and the ＋＋＋ probability was higher; p53 protein expression rate was higher with higher grade of cervical cancer cells, namely worse cancer cells differentiation rate was, the higher the rate of p53 protein was, CIN I compared with CIN II, III, the difference was significant （P 〈 0.05）. Conclusion p53 protein expression is closely related to the CIN occurrence, development,it is early event of cervical cancer.
