摘　　要：目的探讨胃腺癌患者肿瘤组织、癌旁组织中Syk基因的甲基化状态与临床病理特征及预后的关系。方法应用Real—time甲基化特异性聚合酶联反应技术（MSP）检测92例胃腺癌患者肿瘤组织、配对的癌旁组织中Syk基因的甲基化状态，并分析其与临床病理因素及预后之间的关系。选取40例年龄对应的健康志愿者及48例非萎缩性胃炎患者胃镜获取的组织DNA作对照。结果胃腺癌患者肿瘤组织中检测到Syk基因甲基化有33．7％（31／92），明显高于癌旁组织54％（5／92）。健康对照者和胃炎患者均无Syk基因甲基化。Syk基因甲基化与肿瘤大小、生长方式、分化程度、是否有静脉或淋巴管侵犯、TNM分期有关（17〈005）。结论胃腺癌患者Syk基因甲基化与肿瘤的生物学行为有关，提示肿瘤进展，预后不佳。Objective To investigate the aberrant methylation of Syk gene in gastric adenocarcinoma and its clinical sig- nificance. Methods Real-time methylation-specific polymerase chain reaction （qMSP） was used to examine the methylation status of Syk gene in gastric adenocarcinoma tumor tissue and paired pericancerous gastric mucosa of 92 patients with gastric cancer. The relationship of Syk gene methylation and clinicopathologic features and prognosis of patients was analyzed. Forty age-matched healthy subjects and 48 patients with non-atrophic gastritis served as controls. Results The Syk gene methyla- tion was detected in 31/92 （33.7%） cases of gastric adenocarcinoma, which was significantly higher than that in pericancerous mucosa; no methylation was detected in gastric tissue from healthy controls and gastritis patients. Syk gene methylation status were correlated with tumor size, grow pattern, pathological differentiation,TNM stage and invasion of vein and lymphatic vessels （P〈0.05）. Conclusion Ectopic methylation of Syk gene is associated with biological behavior of tumor in gastric adenocarcino- ma, which may indicates tumor progression and poor prognosis.
