摘 要:采用薄层色谱-可见分光光度法以内标法定量测定油脂中甘油三酯的含量。应用硅胶柱层析将混合甘油酯中的甘油三酯分离出来,得到甘油三酯的高纯品,作为甘油三酯测定的标准品。混合甘油酯经薄层色谱分离、洗脱及显色,然后经分光光度法在700nm测定其吸光度,测定甘油三酯的含量。本方法的平均回收率为99.8%,有良好的准确度,精密度略低;获得的甘油三酯标准品,在10~100μg/mL范围内线性关系良好;保证薄层板在5min固定时间显色,洗脱液在1.5h内检测,其稳定性良好;样品的检测限为0.55μg/mL。本方法为混合脂肪酸组成的甘油三酯含量测定提供简单快捷的方法。A method for the determination of triglyceride content in oils and fats was established using thin layer chromatography (TLC) -visible spectrophotometry and internal standard quantification. High-purity triglyceride was separated by silica gel column chromatography from mixed glycerides for use as a reference substance. For the spectrophotometric determination of triglyceride content at 700 nm, triglyceride was separated by TLC before color development. The average recovery rate of the developed method was 99.8%, with high accuracy but slightly lower precision. An excellent linear relationship was observed for the calibration curve established using the prepared triglycedde standard over the concentration range of 10 to 100 μg/mL. Good stability was achieved by color development for 5 min and detection of the eluates within 1.5 h. The limit of detection for triglyceride was 0.55 μg/mL. This method can provide a simple and quick method for the determination of triglyceride conternt.
