摘 要:目的:研究酶解淫羊藿总黄酮制备宝藿苷Ⅰ的工艺。方法:采用β-葡萄糖苷酶酶解淫羊藿总黄酮制备宝藿苷Ⅰ,以宝藿苷Ⅰ得率为指标,通过单因素考察pH值、温度、反应时间及酶与底物质量比对宝藿苷Ⅰ得率的影响。结果:酶解反应的最适条件为温度50℃、pH5.0~5.5、反应时间7 h、酶与底物质量比为1∶20;以淫羊藿总黄酮计,宝藿苷Ⅰ收率为9.25%。结论:β-葡萄糖苷酶酶解淫羊藿总黄酮制备宝藿苷Ⅰ,得率高、工艺简便可靠、反应条件温和,适合工业化生产。OBJECTIVE To study the hydrolysis technology of Epimedium flavonoid by β-glucosidase for preparing baohuo side I. METHODS Taking the yield of baohuoside I as the index, β-glucosidase was used for enzymolysis of Epimedium fla vonoids and the effects of pH value, temperature,reaction time and the ratio of enzyme to substrate on yield of baohuoside I were studied. RESULTS The optimum enzymolysis with β-glucosidase was achieved at 50 ℃, a pH 5.0 - 5.5 Solution for7 h, and the ratio of enzyme to substrate is 1:20. The yield of baohuoside I was 9. 25% (Epimedium flavonoid was computed). CONCLUSION It is feasible to prepare baohuoside I by enzymolysis with β-glucosidase, the condition is moderate and it is suitable for industrialization.
