摘 要:目的:采用蛋白质组学技术,探究中等强度运动后大鼠左室肌蛋白质组的差异表达。方法:将SD大鼠分为运动组和对照组,训练完成后麻醉处死,计量大鼠心重,提取左室肌全蛋白样品,采用2-DE进行分离,选择运动后差异表达量≥10倍的备选目标蛋白点,进行质谱鉴定。结果:取5个目标蛋白点,经质谱鉴定4个。未知蛋白质训练后表达量下调≥12.5倍。结论:1)中等强度运动后,大鼠心脏发生形态学变化使心肌增厚,心收缩力增强;2)训练组大鼠在长期运动训练中,左心室心肌蛋白质组变化显著;3)鉴定出运动医学中还未涉及的4种蛋白质,分别是三磷酸腺苷合酶偶合因子6(CF6)、葡萄糖调节蛋白78(GRP78)、烯酰辅酶A水合酶(ECH)和ATP合酶α亚基(ATP synthase subunit alpha),其表达发生的顺应性改变说明,中等强度运动可提高心肌的能量代谢水平。Objective: Using the technology of proteomics, the study probed into the differentially expressive pro- teome of the left ventricular muscle after medium intensity training. Methods : The researchers divided SD rats into control group and exercise group. After exercise, the rats were killed after anesthesia. The researchers weighed heart of each rat, extracted whole protein samples of the left ventrieular muscle, and separated the protein samples by utilizing two-dimensional gel electrophoresis. The ahernatively objective protein spots whose differential expres- sive volume over lO-fold were selected for MS identification. Resuhs: Taking out five alternatively objective pro- tein spots, the researchers identified four by mass spectrum. The expression volume decreased more than 12.5- folds after unknown protein training. Conclusion : 1 ) The morphologic changes of the rat heart made the cardiac muscle inerassation and strengthened the heart contraction after 8 weeks' medium intensity exercise. 2 ) During long-time training, the rats in the exercise group had significant change in the proteome of the left ventricular mus- cle. 3 )The researchers identified 4 proteins which has not referred in the sports medicine field at present, name- ly, ATP synthase coupling factor 6, glucose-regulated protein 78, Enoyl-CoA hydratase and ATP synthase subunit alpha. The compliance changes indicated that moderate intensity training can improve the energy metabolism level of the cardiac muscle.
