摘 要:目的建立HPLC法检测口服液类保健食品中苯甲酸和山梨酸的含量。方法采用Agilent ZOBAXC18柱(4.6mm×250mm,5μm),柱温为30℃;流动相为甲醇-25mmol.L-1乙酸铵(5∶95);流速为1.0mL.min-1;检测波长为230nm。分别以外标法计算含量。结果苯甲酸浓度在1.209~60.43μg.mL-1范围内呈良好线性关系,r=0.9999;平均回收率为103.8%,RSD为1.7%。山梨酸浓度在1.010~50.51μg.mL-1范围内呈良好线性关系,r=0.9998;平均回收率为95.0%,RSD为1.9%。结论本方法可用于测定口服液类保健食品中苯甲酸、山梨酸的含量,灵敏度高、操作简便、准确可靠。Objective To establish an HPLC method for the determination of benzoic acid and sorbic acid in health food. Methods The Agilent ZOBAX C18 (4.6 mm×250 mm, 5 μm) column was adopted; the column temperature was 30 ℃ and the mobile phase was methanol-25 mmol ·L-1 ammonium acetate (5 : 95)with a flow rate of 1.0 mL ·min^-1, and the detective wavelength was 230 nm. The content was calculated by external standard method. Results A good linear relationship of benzoic acid was found in the range of 1. 209-60.43μg ·mL^-1 (r=0. 9999), and the linear range of sorbic acid was 1. 010-50. 51 μg·mL^-1 (r=0. 9998). The average recoveries were 103.8% and 95.0%, the relative standard deviations (n=9) were 1.7% and 1.9%, respectively. Conclusion The described method can be used for content determination of benzoic acid and sorbic acid in health food, which is simple, sensitive, accurate and reliable.
