摘 要:目的:研究七叶皂苷钠(SE)、碱性成纤维生长因子(bFGF)以及二者联合使用后的保护神经的协同作用,为临床应用提供实验依据。方法:选取64只Wist-ar大鼠,采用改良的Allen的WD法建立急性脊髓损伤模型(胸10平面,25g/cm)模型,随机分为生理盐水组、bFGF组、SE组、SE+bFGF组,每组16只。于术后30分、4、24、48、72小时腹腔注射给药各一次,剂量为生理盐水0.5ml/次,七叶皂苷钠100mg/kg/次,bFGF3600U1次。各组于术后1、3、7、14天分别取4只大鼠处死,应用BBB评分系统和斜板实验、HE染色进行检测、免疫组织化学检测Bcl-2阳性细胞的表达,同时用TUNEL染色法检测神经细胞凋亡,对大鼠进行神经功能评价。结果:与对照组相比,3个治疗组BBB评分、斜板试验及病理学、免疫组织学均优于对照组(P<0.01),并减少了神经元细胞凋亡(P<0.01),改善脊髓损伤后的功能恢复。各个治疗组之间比较,SE联合bFGF组运动功能评分优于其余两组(P<0.01),显著减少了神经元细胞凋亡(P<0.01),保护了损伤后的神经。结论:大鼠脊髓损伤后SE和bFGF均能减轻早期神经功能损伤,减少细胞凋亡,有效保护脊髓神经结构,二者联合使用具有显著的保护神经的协同作用。Objective:To study the aescine sodium (SE),basic fibroblast growth factor(bFGF) and the Protection of neurons in the two joint use of the synergies to provide experimental basis for clinical application.Methods:64 Wistar rats,the Allen WD method to establish the model of acute spinal cord injury(10 flat chest 25g/cm) were randomly divided into 4 groups:saline group,bFGF group,SE group,SE joint bFGF groups;each group is16 rats.The postoperative 30min,4h,24h,48h,72h intraperitoneal injection once a dose:saline solution 0.5ml/times,the aescine sodium 100mg/kg/times of bFGF 3600U time.Each group were collected 3,7 and 14 days after surgery,four rats were sacrificed,the application of the BBB score and the inclined plane test,HE staining were detected,immunohistochemistry to detect Bcl-2 positive cells,using TUNEL staining assay neuronal apoptosis,the evaluation of neurological function in rats.Results:Compared with the control group,the BBB score,the inclined plane test,pathology and immunohistology of the three treatment groups are better than the control group(P<0.01) and reduced neuronal apoptosis(P<0.01),improve function after spinal cord injury recovery;between each treatment group,SE joint bFGF motor function score is better than the other two groups(P<0.01),significantly reduced the neuronal apoptosis(P<0.01),to protect the nerve injury.Conclusion:The results showed that SE and bFGF can reduce early neurological injury after spinal cord injury,decreased apoptosis,the protected the spinal cord structure effectively,both used in conjunction have significant neuroprotective synergy.
