环磷酰胺诱导小鼠HAC肝癌细胞凋亡和抑制增殖的体内实验研究

摘　　要：目的　探讨环磷酰胺对荷瘤鼠HAC肝癌细胞凋亡和增殖的影响。方法　建立小鼠HAC肝癌荷瘤模型 ,用TUNEL法和免疫组化法检测环磷酰胺作用下 ,HAC肝癌细胞的凋亡和PCNA表达的变化。结果　 15mg kg环磷酰胺的抑瘤率 43 %。HAC肝癌细胞的凋亡指数 1 93 % ,较荷瘤对照组 1 11%明显升高 (P <0 0 5 )。增殖指数 35 75% ,较荷瘤对照组 5 6 38%明显下降 (P <0 0 1)。Objective To observe the apoptosis and cell proliferation induced by cyclophosphamide in mice liver cancer. Methods Therapeutic effect of 15 mg/kg cyclophosphamide on mice HAC liver cancer was observed in vivo. Apoptosis and PCNA of tumor cells were also studied by using TUNEL and immunohistochemical methods. Results In the group of control: the apoptotic cell index (AI) in tumor cell was 1.11%. PCNA index (PI) was 56.38 %. In the group of CTX: the rate of tumor inhibition was 43 %; the AI was 1.93 %, represented a significant increase (P<005); the PI was 35.75 %, a statistically significant change (P<001).Conclusion Cyclophosphamide may induce apoptosis and may reduce cell proliferation.$$$$
