摘  要:目的:研究菟丝子黄酮对缺血再灌注损伤模型大鼠脑组织中炎症因子反应的影响。方法:对大鼠以颈内动脉线栓法复制大脑中动脉栓塞脑缺血再灌注模型。实验分为假手术(等容生理盐水)、模型(等容生理盐水)、尼莫地平(12mg/kg)与菟丝子黄酮高、低剂量(100、50mg/kg)组。再灌注结束后进行神经功能缺失症状评分;免疫组化法和Western Blot法检测大鼠脑组织肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、细胞间黏附分子(ICAM)-1的表达。结果:与假手术组比较,模型组大鼠神经功能缺失症状明显,脑组织TNF-α、IL-1β、ICAM-1的表达显著增强(P<0.01);与模型组比较,菟丝子黄酮高、低剂量组大鼠神经功能缺失症状明显改善,TNF-α、IL-1β、ICAM-1的表达显著减弱(P<0.01)。结论:菟丝子黄酮对脑缺血再灌注损伤模型大鼠具有一定保护作用,其机制可能与抑制炎症因子的分泌和表达及减轻脑组织的炎症反应有关。To study the effects of Cuscutae Semen flavonoids on inflammatory response of brain tissue in rats with cerebral ischemia-reperfusion injury. METHODS: Cerebral ischemia-reperfusion injury model of middle cerebral artery occlusion was induced by thread occlusion method of internal carotid. Model rats were divided into sham operation group (constant volume of normal saline), model group (constant volume of normal saline), Nimodipine group (12 mg/kg) group and Cuscutae Semen flavonoids high-dose and low-dose groups (100, 50 mg/kg). Neurological deficits score was conducted at file end of reperfusion; immunohistochemical staining and Western Blotting assay were used to determine the expression of TNF-a, IL-B and ICAM-! in brain tissue of rats. RESULTS: Compared with sham operation group, neurological deficits of rats were improved significantly in model group, and the expressions of TNF-a, IL-Band ICAM-1 in cerebral tissue increased significantly (P〈0.01) ; compared with model group, neurological deficits of rats were improved significantly in Cuscutae Semen flavonoids high-dose and low-dose groups, while the expressions of TNF-a, IL-B and ICAM-1 decreased significantly(P〈0.01). CONCLUSION: Cuscutae Semen flavonoids have a protective effect on cerebral ischemia-reperfusion injury, and its mechanism may be related to inhibition of inflammatorv cvtokine secretion and exnression and reduce the inflammmion af hrmn tissue
