摘 要:依据形态学和生理生化特征,将一株产脂肪酶芽孢杆菌BL03初步鉴定为地衣芽孢杆菌(Bacillus licheniformis)。提取该菌核酸,PCR扩增出地衣芽孢杆菌脂肪酶基因LIP,克隆到pMD19-T载体后进行PCR鉴定及序列测定和生物信息学分析,结果表明该菌LIP基因片段完整,开放阅读框为615bp,编码204个氨基酸,与已发表地衣芽孢杆菌脂肪酶基因(GeneID:3098655)相比,核酸序列相似为99.67%,推导编码氨基酸序列相似性为99.02%,该基因的获得为其异源表达及相关研究奠定了基础。Bacillus strain BL03 was identified as Bacillus lichenifermis based on its morphological, physiology and biochemistry characteristics.The LIP gene encoding lipase was amplified from B.Iicheniformis BL03 and cloned to vector pMD19-T.Nucleotide sequence analysis of the LIP gene revealed that the coding region was formed of one complete ORF including 6].5 base pairs encoding 204 amino acids. The cloned gene was 99.6?% similar with B.licheniformis LIP gene(GenelD:3098655) and the putative amino acid sequence was 99.02% similar respectively. The cloned gene laid foundation for the heterologous expression and relevant researches on it.
