碳酸酐酶-6基因多态性与龋病易感性的初步研究

摘　　要：目的:研究碳酸酐酶-6基因rs2274328(A/C)、rs17032907(C/T)、rs1157666(A/C)3个位点单核苷酸多态性与裕固族和汉族人群龋病易感性的关系。方法:采用病例对照研究,在甘肃裕固族自治县选取两民族样本355例,采集外周静脉血提取DNA,利用核酸质谱技术(MassArray)直接检测3个位点的基因型,应用Haploview4.2以及SPSS16.0软件包进行数据分析。结果:所有基因型分布频率符合Hardy-Weinberg平衡,CA6基因3个位点存在连锁不平衡。rs17032907和rs2274328位点基因型频率在裕固族病例组和对照组中差异有统计学意义(P﹤0.05),其中rs2274328位点CC基因型频率病例组(20.3%)显著高于对照组(5.8%)[OR=3.5,95%CI=1.150-10.649],CA6基因在汉族人群中基因型频率均未见差异有显著性。结论:CA6基因rs2274328位点多态性可能与裕固族人群龋病易感性有关。Objective： To investigate the relationship between the three SNPs of CA6 and dental caries susceptibili- ty in the population of Han and Yugur nationality. Methods： A total of 355 persons in Yugur autonomous counties of Gansu province were chosen and conducted a case-control study. DNA was extracted from peripheral venous blood. The distribution of the genotypes and allele frequency was detected directly by using the method of Sequenom MassArray. The data were statistically analyzed by the Haploview4.2 and SPSS 16.0 software package. Results： All genotypes were in Hardy-Weinberg equilibrium range. A linkage disequilibrium was noted between three SNPs within CA6 gene. Both rs17032907 and rs2274328 SNP genotype frequencies in case and control group of Yugur were statistically significant（P〈0.05）, and the frequency of rs2274328 CC genotype was higher in Yugur nationali- ty case group （20.3%） than those in controls （5.8%） with OR-3.5 （95% confidence interval [CI] =1. 150-10. 649）. However, the differences were not statistical significant in Han. Conclusion： These results suggest that ge- netic polymorphism in the locus of rs2274328 of CA6 gene might be associated with dental caries susceptibility in Yugur population.
