摘　　要：目的　研究部分纯化骨肉瘤血管形成因子并探讨其生物学特性。方法　通过对培养中收集的大量条件培养基进行超滤、层析、透析等截留相对分子质量 8～ 10ku的蛋白质纯化 ,获取 19个层析柱。以人类脐静脉和猪主动脉内皮细胞增殖反应为实验对象 ,用作检验条件培养基和层析产物的生物学活性。结果　条件培养基能显著促进人类内皮细胞和猪主动脉内皮细胞的增殖分别为(336 .91± 49.12 ) /min和 (190 8.33± 2 2 8.38) /min ,层析中的 4～ 6柱 (A值分别为 0 .732± 0 .10 4、0 .80 8± 0 .113、0 .791± 0 .0 91) ,具有显著的促进内皮细胞增殖的生物学效应。结论　人类骨肉瘤细胞能够合成并分泌血管形成因子 ,其相对分子质量在 8～ 10ku之间。Objective To partially purifiyte angiogenesis factor of human osteosarcoma and its biological charaterization. Methods The active peptide with 8～10 ku molecular weight in the conditioned medium obtained from the cultivation of human osteosarcoma cells were partially purified by ultrafiltration, chromatography and dialysis. The angiogenic effects of conditioned medium and the fractions were assessed by proliferation assay of human umbilical vein and pig aorta thoracic endothelial cells.Results The chromatography fractions of 4～6 ( A value:0.732±0.104,0.808±0.113,0.791±0.091 respectively) and the conditioned medium [(336.9±49.12)/min,(1?908.33±228.38)/min] could significantly promote the proliferation of the endothelial cells. Conclusion The human osteosarcoma cells could synthesize and secrete angiogenesis factor with a molecular weight of 8～10 ku.
