摘 要:采用改良的CTAB法提取来自不同地区的9种中药鸡骨草的总DNA,利用豆科植物通用引物对matK序列进行PCR扩增,将扩增得到的序列提交到GeneBank中获得登录号。最后将所有序列进行聚类分析,计算种问及种内遗传距离.建立系统发育邻接树。结果表明,marK序列的全长为889-895bp;不同植物之间的遗传距离较大,同种植物之间的遗传距离较小,且最大的种内遗传距离小于最小的种间遗传距离。因此,matK序列可以作为豆科植物的DNA条形码。We got the DNA from nine different Abrus cantoniensis which grow in Guangdong and Guangxi province by using the modified CTAB method. In order to get nine kinds of Abrus cantoniensis ITS sequences, we amplified the rDNA internal transcribed spacer region (ITS) sequences by using universal primers through PCR, as well as registered to GenBank and got tile accession number. Finally, all sequences were analyzed using the cluster analysis to calculate interspecific and intraspecific genetic distance for establishing phylogenetic adjacent tree. Tile results showed that the total length of the matK sequence was 889-895 bp. Genetic distance calculation results showed that the genetic distance between different plant species was large, and the genetic distance between the same plant species was small. However, the maximum intraspecific genetic distance was smaller than the minimum interspecific genetic distance. Therefore, the matK sequence could be used as DNA barcode of Fabaceae.
