摘  要:目的:采用细胞培养方法研究顺铂不同给药方式对鼻咽高分化鳞癌细胞株CNE-1的细胞毒性以及细胞周期的影响。方法:MTT方法检测顺铂对鼻咽癌CNE-1细胞的半抑制浓度(IC50),1/5IC50为小剂量化疗参考剂量,而IC70则为MTD化疗参考剂量。细胞分4组进行如下处理:A组剂量为1/5IC50的顺铂连续作用96 h;B组剂量为IC70的顺铂连续作用3 h;C组为IC70作用3 h后间隔93 h;D组为未加入顺铂化疗的鼻咽癌细胞株同样条件下培养96 h。采用流式细胞仪进行细胞周期的检测。结果:顺铂对鼻咽癌CNE-1细胞株作用72 h的IC50值为0.26μg/ml,1/5IC50为0.05μg/ml,IC70为0.72μg/ml。顺铂为0.05μg/ml剂量时对CNE-1细胞无明显毒性,但细胞周期研究发现,当其连续作用96 h时,与阴性对照组相比可引起明显G2/M期阻滞(P<0.05);而顺铂剂量为0.72μg/ml连续作用3 h后间隔24 h以及93 h时检测细胞周期变化也出现显著G2/M期阻滞(P<0.05);但在0.72μg/ml的顺铂作用3 h后立即检测细胞周期变化,与相应阴性对照组相比无统计学意义(P>0.05)。结论:体外低毒性的小剂量顺铂连续给药可诱导鼻咽癌细胞株CNE-1细胞周期G2/M期阻滞。Objective:To study the effect of different treatment protocols of cisplatin on cytotoxicity and cell cycle in well-differentiated nasopharyngeal squamous cell carcinoma cell line CNE-I. Methods:MTT assay was used to evaluate the half inhibitory concentration (IC50) of cisplatin. The reference dose of low-dose chemotherapy was 1/5IC50, and IC70 as MTD reference dose. Nasopharyngeal carcinoma cells were treated with cisplatin by four treatment protocols: 96h exposure to dose of 1/5IC50 (protocol A); 3h exposure to dose of IC70 (protocol B); 3h exposure to IC70 followed by a 93h interval(protocol C);cells not exposed to cisplatin were cultured under the same conditions for 96h (protocol D).The change of cell cycle were studied by flow cytometry. Results: The IC50 of 72h exposure to cisplatin was 0.26 μg/ml , and the dose of 1/5IC50 and lCTo was 0.05 μg/ml and 0.72μg/ml, respectively. It had no significant toxicity when the dose of cisplatin was 0.05 μg/ml, however the cell cycle study found that compared with the negative control group G2/M phase was significantly arrested after 96h exposure to this dose (P〈0.05),3h expose to 0.72 μg/ml of cisplatin with a 24h interval and 93h interval could induce significant G2/M phase arrest (P〈0.05), whereas no cell cycle arrest was found immediately after 3h exposure to 0.72 μg/ml of cisplatin (P〉0.05). Conclusion:The protocol of low-dose continuous administration of cisplatin, which was hypotoxic, could induce G2/M phase arrest in vitro study.
