摘 要:目的探讨哌甲酯对注意缺陷多动障碍动物模型——自发性高血压大鼠(SHR)在定位航行实验中搜索策略的影响。方法实验分三组,16只SHR随机分为模型组和哌甲酯组,每组8只,韦氏塔京都(WKY)大鼠8只为对照组。应用Morris水迷宫对三组大鼠分别进行6 d定位航行测试。每天实验前半小时,哌甲酯组腹腔注射干预药物10 mg/kg,另外两组给予等体积生理盐水。结果 (1)自训练第2天起,各组使用有效搜索策略寻找隐藏平台的频率均较第1天有所提高(P<0.05)。(2)哌甲酯组在训练第1,3,6天较模型组选择了更多的直线式搜索(P<0.01)。结论哌甲酯可能通过优化SHR水迷宫搜索策略,尤其是增加高效率的直线式搜索来提高其学习能力。Objective To investigate the influence of methylphenidate on search strategy of spontaneously hypertensive rat (SHR) in place navigation test. Methods The SHR rats were randomly divided into model group (8 rats) and methylphenidate hydrochloride group (MPD group, 8 rats), and 8 Wistar Kyoto rats were defined as control group. Rats in the 3 groups were tested with place navigation on 6 consecutive days. Rats in MPD group were injected intraperitoneally with 10mg/kg methylphenidate hydrochloride 30 minutes before place navigation test, while rats in model group and control group were injected equal-volume saline in the same way. Results Compared with the performance on day 1, the frequency of using effective straight strategy to search the hidden platform increased significantly in all the 3 groups from the second day of training (P 〈 0.05 ). Additionally, rats in MPD group used straight strategy more frequently than rats in model group on day 1, day 3 and day 6 respectively ( P 〈 0.01 ). Conclusion Methylphenidate can improve the learning ability of SHR rats by optimizing the Morris water maze search strategy, especially by using orthoscopic search strategy.
