金星无核葡萄胚挽救影响因素的研究

摘　　要：以金星无核葡萄的胚珠为试材,对胚挽救的多个影响因素包括接种时间、培养基和培养方式进行研究,以进一步确定无核葡萄胚挽救各个阶段的适宜培养条件。结果表明,影响无核葡萄胚挽救最重要的因素是接种时期,其他因素影响程度因组合而不同:在果粒着色10%左右接种胚珠发育率最高,达到63.38%;在胚珠发育阶段,附加1.5 mg/L IBA、1.0 mg/L 6-BA和0.5 mg/LGA3的ER培养基最适宜离体幼胚的发育;在胚萌发阶段,使用附加1.0 mg/L IBA、0.5 mg/L 6-BA和0.5 mg/L GA3的WP培养基萌发率最高,同时对胚珠进行横切处理可以显著提高胚珠萌发出苗率。The open pollination ovules of Venus Seedless Grape were used as tested materials, and the effect of inoculating time, culture media and ovule processing method on embryo rescue of Venus seedless grape were studied to determine the suitable culture conditions of em- bryo rescue in various stages. The results showed that the most important factor affecting the embryo rescue of seedless grape was the inocula- ting time, and the influeneing level of other factors were different among combinations ： the suitable period of inoculation was coloring in fruit peel of 10 %, the highest growth rate reached 63.38 % ; ER medium supplemented with 1.5 mg/L IBA, 1.0 mg/L 6-BA and 0.5 mg/L GA3 were found to be the most suitable for embryo development; the suitable medium for embryo germination was WP ＋ 1.0 mg/L IBA ＋ 0.5 mg/L 6-BA ＋ 0.5 mg/L GA3 ,meanwhile it also showed that the ovule by crosscut significantly increased the germination rate.
