摘 要:以矮牡丹花粉为试材,利用离体培养法对其花粉生活力进行了测定,研究了不同贮藏条件对花粉萌发的影响以及花粉形态与萌发之间的关系。结果表明:培养基中添加蔗糖和硼酸对花粉的萌发有促进作用,矮牡丹花粉萌发培养基中最适宜的蔗糖和硼酸浓度分别为:9%、0.0045%,矮牡丹花粉萌发率达58.52%。花粉生活力随贮藏时间的延长而下降,不同的贮藏条件对牡丹花粉生活力有显著的影响,矮牡丹花粉最佳贮藏方法为-196℃超低温保存,耐贮藏力超过378 d。花粉形态与其萌发之间有相关性。Taken the fresh pollens of dwarf peony as tested material, the fresh pollen ability was determined with the method of germination in vitro, the effects of different storages on the pollen germination and the relation between pollen morphology and pollen germination were studied. The results showed that culture media with different concentration of sucrose and boric had good effects on the pollen germination. The germinating percentage of dwarf peony pollen reached the highest after incubated on the media supplemented with 9 % sucrose and 0. 0045 % boric acid, and was 58.52 %. Vitality of pollen were gradually decreased with the passage of storage time and difference under en- virenments, the temperature of the best environment to keep dwarf pollen vitality was at - 196℃, and its storage time was over 378 d. There was a correlation between pollen morphology and pollen germination.
