摘 要:研究血桐叶提取物的抑菌及抗氧化活性,为血桐的开发利用提供科学依据。首先以70%乙醇回流提取血桐叶,依次用不同有机溶剂萃取,得到石油醚、乙酸乙酸、正丁醇萃取部位和水层部位;然后采用滤纸片法与最低抑制浓度法,考察血桐醇提取物各萃取部位对5种实验菌的抑菌效果;采用福林一酚法、DPPH法和邻苯三酚法分别测定血桐各萃取物的总多酚含量及其对自由基和超氧阴离子(.O2-)的清除作用。结果表明:血桐石油醚、乙酸乙酯、正丁醇萃取物对金色葡萄球菌、白色葡萄球菌、藤黄微球菌均具有较强的抑菌效果,其最低抑制浓度(MIC)为0.2~0.6mg/mL,对大肠杆菌、枯草芽胞杆菌的抑制作用均较弱。石油醚、乙酸乙酯、正丁醇及水层萃取物4个部位的酚性含量分别为16.35%、30.38%、50.73%、24.64%。DPPH法与邻苯三酚法测定血桐醇提物不同萃取部位的抗氧化能力大小顺序为正丁醇萃取物>乙酸乙酯萃取物>水层部位>石油醚萃取物。血桐叶提取物具有较强的抑菌及抗氧化活性,为进一步研究其抑菌和抗氧化机理提供了科学依据。Antimicrobial and antioxidant activities of the extracts of Macaranga tanarius leaves were investigated.The leaves were extracted with 70%(v/v) EtOH.After the solvent was evaporated,the resultant extract was then extracted in turn with petroleum ether,ethyl acetate,n-BuOH,and water.The antibacterial activities of the extracts were measured by using filter paper method and minimum inhibiting concentration method.The contents of phenols in the extracts were measured by Fulin method,and the capabilities of scavenging free radical and super oxide anion of the extracts were evaluated by DPPH method and 1,10-phenanthroline-Fe2+ oxidation method.The extracts from petroleum ether,ethyl acetate,and n-BuOH exhibited higher inhibition effect to Staphylococcus aureus,S.albus,Micrococcus luteus than other strains,with a MIC range of 0.2 to 0.6 mg/mL.The contents of phenols were 16.35%,30.38%,50.73%,and 24.64%,respectively in the extracts from petroleum ether,ethyl acetate,n-butanol,and water.The antioxidant activities of the extracts were correlated to the contents of total phenols.The results demonstrated that the alcoholic extracts from M.tanarius leaf possessed good antibacterial and antioxidant activity,which provided scientific basis for the research of the mechanism of its bioactive activities.
