摘  要:目的:观测高脂饮食诱导雌性肥胖大鼠血糖、胰岛素、血脂及肾周脂肪组织(PAT)与皮下脂肪组织(SAT)内脏脂肪组织特异性丝氨酸蛋白酶抑制剂(Vaspin)mRNA的变化,探讨Vaspin在雌性肥胖大鼠糖脂代谢中的作用。方法:断乳雌性大鼠随机分成普通饮食组(A组)与高脂饮食组(B组),分别给予普通饲料与高脂饲料。实验开始时及喂养4周后测量体重(W)、体长(H)及腹围(AC)。取血并取SAT与PAT。氧化酶法测定空腹血糖(FPG),放射免疫法测定空腹胰岛素(FINS),酶法测定血总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)与高密度脂蛋白(HDL),实时定量聚合酶链反应(Real-time PCR)测定Vaspin mRNA水平。结果:两组大鼠W、H及AC差异均有统计学意义;B组大鼠FPG、FINS、TC、TG、LDL及PAT Vaspin mRNA水平均高于A组,HDL水平低于A组,SAT Vaspin mRNA水平差异无统计学意义。结论:高脂饮食诱导的雌性肥胖大鼠模型建立成功,出现糖脂代谢紊乱;PAT与SAT均可检测到Vaspin mRNA表达;雌性肥胖大鼠PAT Vaspin mRNA高于雌性非肥胖大鼠。Objective : To investigate the effects of high-fat diet induced female obese rats on glucose, insulin, lipid and Vaspin mRNA profiles of visceral adipose tissue(VAT) and perire'nal adipose tissue(PAT) and to speculate effects of Vaspin on glucose and lipid metabolism. Methods:Female weaned rats were randomized into normal diet group(group A) and high-fat diet group(group B), to which corresponding diet were given separately. The weight(W), height(H) and abdominal eircumference(AC) were measured before and after 4 weeks of special diet and blood samples, VAT and PAT were harvested. FPG levels were detected by glucose oxidase method, FINS lev- els were detected by radioimmunoassay method, total cholesterol(TC), triglyceride(TG), low density lipoprotein (LDL), high density lipoprotein(HDL) levels were detected by glycerol phosphate oxidase-peroxidase method, Vaspin mRNA levels were measured by real time polymerase chain reaction(Real-time PCR). Results: After 4 weeks of special diet, there were statistical differences of W,H and AC between the 2 groups. FPG, FINS, TC, TG, LDL and PAT Vaspin mRNA levels of group B were higher than group A while HDL levels of of group B were lower than group A, there was no statistical difference of SAT Vaspin mRNA levels between the 2 groups. Conclusion;High-fat diet induces obesity in female rats and metabolic disturbance of glucose and lipid;Vaspin mRNA can be detected in both VAT and PAT in female rats; Vaspin mRNA profile in PAT in female obese rats is higher than the one in female non-obese rats.
