摘  要:目的:通过建立裸鼠子宫内膜癌移植瘤模型,观察G蛋白偶联受体抑制剂百日咳毒素(PTX)对子宫内膜癌移植瘤细胞凋亡及对磷脂酰肌醇3激酶/蛋白激酶B(PI3K/Akt)信号传导通路中磷酸化蛋白激酶B(p-Akt)表达的影响。方法:将体外培养的人子宫内膜癌HEC-1A、Ishikawa细胞系,分别接种于28只裸鼠右背部皮下,建立裸鼠子宫内膜癌移植瘤模型,每个细胞系随机分为2组,对照组(腹腔注射生理盐水)和实验组(腹腔注射PTX),绘制肿瘤生长曲线,计算移植瘤的体积、抑瘤率等。光学显微镜下观察移植瘤组织的病理学特点,Tunel技术检测移植瘤组织中细胞凋亡的情况,免疫组织化学检测p-Akt的表达。结果:(1)PTX均能有效抑制HEC-1A、Ishikawa细胞裸鼠移植瘤生长,抑瘤率分别为38.74%、33.21%。(2)HE染色显示,实验组细胞异型性、核浆比相对较小;Tunel染色显示,HEC-1A、Ishikawa细胞系实验组细胞凋亡率均显著高于对照组(P均<0.05)。(3)免疫组织化学结果显示,PTX能降低p-Akt的表达,HEC-1A实验组与对照组比较差异显著(P<0.05),Ishikawa实验组与对照组比较差异无统计学意义(P>0.05)。结论:PTX可能通过抑制GPR30及其介导的PI3K/Akt通路,抑制子宫内膜癌裸鼠移植瘤的生长,促进其凋亡。Objective: To investigate the inhibition effects of PTX and the effects of p- Akt expression on xenograft tumors of human endometrial carcinoma HEC-1A, Ishikawa ceils in nude mice. Methods: The models of xenograft tumors were established by subcutaneous inocula- tion. The effects of 0.9% NaC1, PTX on xenograft tumors were observed. The anti-tumor efficacy was evaluated by measuring tumor volume and tumor growth. The histopathological changes of tumor specimens were observed by HE staining. Cell apoptosis and the expression levels of phosphorylated-Akt(p-Akt) in the xenograft tumors were detected by tunel and immunohisto- chemistry respectively. Results: ( 1 ) PTX can significantly supress the growth of endometrial cancer xenografs. The inhibition tumor rates were 38.74% and 33.21% respectively. (2)HE staining showed that the proportion of nucleus and cytoplasm was smaller and have lower density of vessels. Tunel assay showed a higher apoptosis rate in the experimental groups (P〈O. 05 ). (3) Immunohistochemistry showed that PTX can effectively reduce the expression of p-Akt in tumors (HEC-1A,P〈0.05 ;Ishikawa,P〉0.05 ). Conclusions: PTX can decrease the xenograft tumors growth and promote the apoptosis of endometrial carcinoma HEC-1A,Ishikawa cells in vivo through blockage PI3 K/Akt signaling pathways.
