Objective: To examine the sex differences in the development of experimental neuropathic pain after partial sciatic nerve injury in the rats. Methods: Spared sciatic nerve injury model in adult rats has been established. The injury-induced hypersensitivity to mechanical and cold stimulation (mechanical and cold allodynia) has been studied and sex difference has been examined. Results: Spared sciatic nerve injury produced mechanical and cold hypersensitivity of the ipsilateral hindpaw in rats. Although the magnitude of mechanical hypersensitivity was similar between the two sexes, the hypersensitivity in females occurred earlier (female 1 day, male 3 days) and was more prolonged than males (female 84 days, male 49 days). The cold hypersensitivity was significantly more severe in females than in male rats (P 〈 0.01). Conclusion: Rats exhibited neuropathic pain-like behaviors after spared sciatic nerve injury in the forms of mechanical and cold allodynia. There are significant sex differences with female rats developing more severe mechanical and cold hypersensitivity compared with the male rats.
