摘 要:目的观察FSCN1在人乳腺癌及乳腺良性病变组织中的表达及其对人乳腺癌细胞增殖的影响。方法采用免疫组化MaxVision两步法检测124例乳腺癌和27例乳腺良性病变组织中FSCN1蛋白的表达。用干扰RNA技术沉默人乳腺癌细胞株MCF-7中FSCN1基因的表达,通过MTT实验检测细胞增殖能力。结果 FSCN1蛋白在乳腺癌和乳腺良性病变组织中的阳性率分别为37.9%(47/124)和51.9%(14/27),差异无统计学意义(P>0.05);FSCN1蛋白阳性率随组织学分级的增高而增高(P<0.01);ER、PR阴性乳腺癌患者的FSCN1蛋白阳性率明显高于ER、PR阳性患者(P<0.05);转染FSCN1 siRNA后,MCF-7细胞的增殖活性较siRNA NC组明显降低(P<0.05)。结论 FSCN1对人乳腺癌细胞的增殖能力可能存在正性调控作用,有望成为乳腺癌潜在生物学标记物。Purpose To investigate the expression of FSCN1 protein in breast carcinoma and its effect on breast cancer cell prolifera- tion. Methods One hundred and twenty-four cases of breast carcinoma and 27 cases of benign breast lesions were analyzed by immu- nohistochemical staining. RNA interference was used to inhibit FSCN1 expression in human breast cancer line MCF-7. Cell prolifera- tion was evaluated by MTr assay. Results FSCN1 protein expression rates in breast cancer tissue and benign breast lesions were 37.9 % (47/124) and 51.9 % ( 14/27 ), respectively (P 〉 0. 05 ). FSCN1 protein expression increased with the increasing of histolog- ical grading of the cancer (P 〈 0. 01 ). FSCN1 protein positive rates in tumors with negative ER and PR status were significantly higher than those of ER and PR positive patients (P 〈 0. 05). Transfection of FSCN1 siRNA into MCF-7 cells led to a significant decrease in cell proliferation compared to the siRNA NC transfectants (P 〈 0. 05). Conclusion FSCN1 might positively regulate proliferation of breast cancer cells and might be a potential candidate of biological biomarker for breast cancer.
