摘 要:为了探讨鲎素对细菌基因组DNA和RNA作用的分子机制,采用琼脂糖凝胶阻滞电泳、荧光和紫外光谱的方法研究了鲎素对细菌基因组DNA和RNA的影响,结果表明鲎素能够与大肠杆菌基因组DNA发生作用,并呈浓度依赖关系,浓度越高对细菌基因组DNA损伤越大;鲎素也能与基因组DNA和RNA结合,抑制其迁移。鲎素与基因组DNA的结合方式有待进一步研究,此结果为在分子水平上深入了解鲎素的杀菌机制提供重要的理论依据。In order to investigate the molecular mechanism of tachyplcsin I role on bacterial genome, the genomic DNA and RNA binding ability of tachyplesin I was examined by agarose gel retardation eleetrophoresis, fluorescence and UV spectroscopy in this experiment. The results indicated that tachyplesin I had effect on genomic DNA in a concentration-dependent manner, and the high concentrations of tachyplesin I ( above or equal to 80 p^g/mL) could lead to genomic DNA breaking. Tachyplesin I could combine with genomic DNA and RNA, and inhibit their migration. The mode of combining taehyplesin I and genomic DNA needs further study. The results provided a theoretical basis for further understanding the sterilization mechanism of tachyplesin I at molecular level.
