摘 要:目的:观察二甲双胍联合阿霉素应用对人乳腺癌细胞MDA-MB-231增殖和凋亡的影响。方法:MTT法分别检测二甲双胍、阿霉素和二甲双胍联合阿霉素对MDA-MB-231细胞生长的抑制作用;平板克隆实验检测二甲双胍联合阿霉素对MDA-MB-231细胞克隆形成能力的影响;流式细胞仪检测二甲双胍联合阿霉素对MDA-MB-231细胞凋亡的影响。结果:二甲双胍和阿霉素分别对MDA-MB-231细胞生长有抑制作用,二甲双胍联合阿霉素应用对MDA-MB-231细胞生长的抑制作用更加显著,并且随着药物浓度的增加而增加;二甲双胍联合阿霉素应用与单药相比能够明显降低MDA-MB-231细胞克隆形成率,并且促进细胞凋亡。结论:二甲双胍联合阿霉素应用与单药相比能够显著抑制人乳腺癌细胞MDA-MB-231细胞的增值,促进其凋亡,可见两药联用对肿瘤细胞的杀伤具有协同性。Objective: To investigate the effect of combining metformin and Adriamycin on the proliferation and apoptosis effect of MDA-MB-231 cell line. Methods: MDA-MB-231 cells were treated by metformin and Adriamycin for 4 days. The proliferation of MDA-MB-231 was tested by MTT assay. The clone formation of MDA-MB-231 was assessed by clone formation assay. The cell apoptosis was detected by FCM. Results: Metformin or Adriamycin could inhibit MDA-MB-231 cells proliferation, respectively. Moreover, after combination of two drugs the cell proliferation was inhibited more seriously. Combination of two drugs could inhibit cell clone formation and promote cell apoptosis significantly. Conclusion: Metformin and Adriamycin can inhibit MDA-MB-231 cells proliferation and clone formation and promote cell apoptosis effectively.
