摘 要:目的:探讨乌骨藤制剂对人肝癌细胞Bel-7402株的影响并初步探讨其机制。方法:分别用不同浓度的乌骨藤制剂处理人肝癌细胞Bel-7402,未经药物处理的细胞为阴性对照组,采用MTT法检测肝癌Bel-7402细胞增殖、流式细胞术检测人肝癌细胞Bel-7402的凋亡情况以及p53蛋白的表达。结果:不同浓度的乌骨藤制剂(10mg/ml,20mg/ml,40mg/ml)处理肝癌Bel-7402细胞后,其增殖受到抑制,作用24h的抑制率分别为11.57%、21.09%、31.27%,48h的抑制率为27.55%、35.95%、45.13%,随着药物浓度的增加(20mg/ml,40mg/ml,80mg/ml)细胞凋亡显著增加,凋亡率分别为(12.2±0.67)%、(17.4±0.69)%、(22.9±1.38)%,而p53蛋白的表达也随着药物浓度的增加而增加。结论:乌骨藤制剂能够抑制人肝癌细胞的增殖,诱导其凋亡,作用机制可能与激活p53基因有关。Objective:To study the effect of Glaucescent Fissistigma Root preparation on Be1-7402 cells and its mechanisms. Methods:After being treated by Glaucescent Fissistigma Root injection of different concentrations, untre- aed cells as control, viability of Bel - 7402 cells was determined by MTI method, apoptosis and p53 protein assessed by FACS. Results:The inhibition( 11.57% ,21.09% and 31.27% in 24h,27.55% ,35.95% and 45.13% in 48h) was observed while the cells were treated by different concentrations of Glaucescent Fissistigma Root injection. Also, the apoptosis [ ( 12.2 ±0.67) %, ( 17.4 +0.69) % (22.9 ±1.38) % ] and the increasing of p53 protein was detec- ted 'after treated by the injection(20mg/m1,40mg/ml,80mg/ml). Conclusion:Glaucescent Fissistigma Root injection could inhibit the proliferation of Bel -7402 cells and induce apoptosis,the mechanisms may be related with p53 gene being activated.
