摘 要:目的:研究铜绿假单胞菌注射液(PA-MSHA vaccine PA)对肝癌瘤株细胞的体外杀伤作用,并观察其对肝癌瘤株细胞的体外增殖影响及杀伤作用的剂量依赖性。方法:采用MTT比色法观察不同剂量PA对肝癌瘤株细胞生长抑制作用和杀伤作用的影响;扫描电镜观察PA与肝癌瘤株细胞的结合过程及肝癌瘤株细胞的凋亡过程;双染色流式细胞术观察不同剂量铜绿假单胞菌注射液对肝癌瘤株细胞的凋亡影响。结果:PA能结合肝癌瘤株并进入细胞内部,诱导细胞凋亡,从而抑制肝癌瘤株细胞增殖和生长,产生杀伤作用,且杀伤作用随着用药浓度增加而加强。结论:铜绿假单胞菌注射液有直接杀伤肝癌瘤株细胞的作用,且这种作用存在剂量依赖性。Objective.To investigate the killing effect by PA -MSHA vaccine on hepatocarcinoma cells in vitro, and observe the dose - dependent inhibition and killing effect by PA - MSHA vaccine on hepatocarcinoma cell. Meth- ods :The MTY olorimetric method was used to observe the inhibition and killing effect by different doses of PA - MSHA on hepatocarcinoma cells;Combination process of PA - MSHA with hepatocarcinoma cells and apoptosis of hepatocarcinoma cells were observed by scanning electron microscopy;Apoptosis process of hepatocarcinoma cells by different doses of PA - MSHA was observered by double dyeing flow cytometry art. Results: PA - MSHA vaccine could penetrate into the hepatocarcinoma cells so as to inhibit its proliferation and the killing effect was in direct proportion to its dose. PA - MSHA vaccine could induce hepatocarcinoma cells apoptosis. Conclusion: PA - MSHA vaccine has killing effect on hepatocarcinoma cells and the effect is dose - dependent.
