摘 要:目的:研究HepG2细胞对黄连生物碱的吸收及黄连生物碱对线粒体复合物Ⅲ表达的影响。方法:传代培养HepG2细胞,将其分为对照组、药根碱组、表小檗碱组、黄连碱组、巴马汀组、小檗碱组,设置每组药物浓度由低到高(0.25、0.50、1.00、2.00、4.00μg/mL),培养24h。通过HPLC法和Western blot法分别检测各组细胞中黄连生物碱的含量及线粒体复合物Ⅲ的含量。结果:在同一浓度下,HepG2细胞中小檗碱的浓度最高,黄连碱次之,药根碱则不能吸收;细胞中的线粒体复合物Ⅲ含量,小檗碱组最高,且小檗碱含量与线粒体复合物Ⅲ含量负相关;黄连碱组含量次之,且随着浓度变化不大;表小檗碱组、巴马汀组则影响不大。结论:小檗碱和黄连碱能提高线粒体复合物Ⅲ的表达,其他生物碱则无明显影响。Objective:The purpose was to study the absorption of alkaloids from Rhizoma Coptis by HepG2 cells and their effect on the expression of Mitochondrial Complex Ⅲ. Methods= HepG2 cells were grouped by control, jatrorrhizine, protoberberine, coptisine, paimatine and berberine, with concentrations from high to low (0.25,0.50, 1.00,2.00,4.00μg/mL)for 24h.HPLC was used to detect concentration of alkaloids in cellular,and Western blot was used to study alkaloids' function on Mitochondrial Complex Ⅲ. Results: It was found that at the same concentration, HepG2 cells could absorb berberine more easily, followed by coptisine,jatrorrhizine could not be absorbed. Besides, berberine and coptisine could enhance the expression of Mitochondrial Complex Ⅲ. Conclusion:The experiment showed that berberine and coptisine could cure Diabetes and Diabetic complications, while other alkaloids effect slightly.
