摘 要:目的:以小肠黏膜下层(SIS)吸附磁颗粒作为热疗介质,研究靶向磁热疗对小鼠结肠癌皮下移植模型的作用。方法:选择Balb/c雌性小鼠66只,4~6周龄,体质量22~25g,CT26结肠癌细胞皮下注射建立小鼠模型。将磁流体颗粒沉淀于置备好的SIS上,手术植入实验组小鼠结肠癌组织周围进行包裹。每24h将实验组小鼠暴露在突变磁场下60min,共3次。对应设置空白对照组,SIS对照组,磁流体对照组,磁场对照组进行对比研究,每组11只。观察各组小鼠的肿瘤生长情况和小鼠生存时间,检测各组小鼠各主要脏器的病理变化。结果:磁热疗作用后,实验组肿瘤中心温度在15min后稳定在43℃,而正常组织温度则始终稳定在24℃。病理检测显示,各组小鼠各主要脏器未见明显结构异常。与各对照组相比较,实验组小鼠肿瘤生长受到显著抑制,P<0.05;生存时间显著延长,P<0.05。结论:以SIS吸附磁颗粒作为热疗介质的靶向磁热疗能显著抑制小鼠结肠癌模型的肿瘤生长,延长小鼠生存时间。靶向磁热疗的进一步研究有可为结肠癌治疗提供新的方法。OBJECTIVE:To study the anticancer effect of targeted hyperthermia in subcutaneous mouse models of co- lon cancer,using small intestinal submucosa (SIS) combined magneticnanoparcicles as the hyperthermia medium. METH- ODS: Totally 66 Balb/c female mice were selected,4--6 weeks old,22--25 g weight. Mouse models of colon cancer was established by injecting CT26 cells subcutaneously. SIS combined magneticnanoparcicles were implanted around the tumor tissue of the mice in the trial group. These mice were then placed in alternating magnetic field for 60 minutes every 24 hours for three times. Correspondently,a bland control group,a SIS group,a magneticnanoparcicles group and a magnetic field group were established as control groups with 11 mice in each group. The tumor growth and mice survival in each group were recorded and the pathological changes in principal organs were detected. RESULTS: After the mice in trial group were exposed to the alternating magnetic field,the central temperature of the tumor tissue ascended to 43℃ fifteen minutes later,whereas the temperature in the normal tissue were kept in 24 ℃. No obvious pathological abnormality in principal organs of each group was found. Compared with each control group,tumor growth in the trial group was appar- ently inhibited (P〈0.05) and the mice survival time in this group was prolonged (P〈0.05). CONCLUSIONS: Using SIS combined magneticnanoparcicles as the hyperthermia medium,targeted hyperthermia can obviously restrain the growth of colon cancer in the subcutaneous mouse tumor models and prolongs the survival of nude mice. Further studies in this field may provide new approaches to the treatment of colon caner.
