摘 要:目的测定不同地区香薷各部位总黄酮和游离黄酮的含有量,找出含黄酮量较高的药材产地和黄酮的主要储存部位。方法以紫外可见分光光度法(UV-Vis)测定香薷总黄酮,最适条件是0.05 mol/L三氯化铝,pH值为5.0的乙酸缓冲液,水浴温度50℃,水浴时间30 min,检测波长420 nm。采用反相高效液相色谱法测定3种主要游离黄酮,色谱柱为Welchrom TM C18(200 mm×4.6 mm,5μm),以0.4%磷酸水溶液(A)-甲醇(B)为流动相,梯度洗脱;体积流量1.0 mL/min;检测波长350 nm;柱温25℃。结果木犀草素、芹菜素和黄芩素-7-甲醚分别在0.88~176μg/mL、1.1~219.2μg/mL和1.55~149.4μg/mL范围内表现出良好的线性(r>0.999);方法的回收率在98.80%~102.01%之间,RSD均小于5%,精密度和稳定性良好。以江西和湖北产的香薷总黄酮含有量较高,其全草中总黄酮的质量分数分别为5 955.35μg/g和4 362.28μg/g,特别是江西和湖北产香薷花苞中总黄酮的质量分数分别高达13 335.99μg/g和12 936.46μg/g。结论不同地区香薷总黄酮含有量由高到低依次为江西、湖北、河南、浙江,不同部位总黄酮的含有量由高到低依次为苞、尖、茎、根。江西产香薷的总黄酮和3种主要游离黄酮的量均较高,且主要集中在茎以上部位。AIM To determine the contents of total and free flavonoids from different tissues of Mosla chinensis in different habitats,and to select and find the raw materials with higher contents and main storage sites of the flavonoids.METHODS Total flavonoids in Mosla chinensis were determined by UV spectrophotometer,and the optimized analysis conditions consisted of concentration of aluminum chloride(0.05 mol/L),pH 5.0 of acetate buffer with water bath at 50 ℃ and 30 min in detection wavelength of 420 nm.Free flavonoids(luteolin,apigenin and negletein) were measured by RP-HPLC with Welchrom TMC18(200 mm×4.6 mm,5 μm) column and mobile phase(0.4% phosphoric acid-methanol) in gradient elution mode at 1.0 mL/min flow velocity,350 nm detection wavelength and 25 ℃column temperature.RESULTS The results showed that the contents of luteolin,apigenin and negletein were good linearity and the correlation coefficients(r>0.999) in 0.88-176 μg/mL,1.1-219.2 μg/mL and 1.55-149.4 μg/mL,respectively.The recoveries were 98.80%-102.01% with less than 5% RSDs,and the higher contents(5 955.35 μg/g and 4 362.28 μg/g) were respectively presented in the samples from Jiangxi and Hubei Province.Particularly,the highest contents(13 335.99 μg/g and 12 936.46 μg/g) were determined in the bud part of Mosla chinensis from above two provinces.CONCLUSION The study shows that the contents of total flavonoids in Mosla chinensis from different habitats are Jiangxi,Hubei,Henan,Zhejiang Province in descending order,and in different parts are flower bud,branchlet,stem,root in descending order.The contents of total flavonoids and mainly three free flavonoids in Mosla chinensis from Jiangxi Province are higher than those from other provinces,and they are mainly enriched in above parts of stem.
