摘  要:目的探讨杀伤细胞免疫球蛋白样受体(killer cell immunoglobulin—like receptor,KIR)2D和人类白细胞抗原Cw(human leucocyte antigenCw,HLA—Cw)基因型组合与子痫前期发病的关系。方法采用PCR一序列特异性引物(PCR—sequence specific primer,PCR—SSP)方法对117例子痫前期孕妇(子痫前期组)和106例正常健康孕妇(正常晚孕组)及其新生儿进行KIR2D、HLA—Cw基因分型,比较其基因频率,并对其配对进行比较分析。结果单个基因型检测发现KIR2DS2和HLA—Cwl2基因型分布在子痫前期组(5.12%和8.12%)显著高于正常晚孕组(1.58%和3.30%),差异均有统计学意义(P均〈0.05):且在KIR2D基因亚型组合分析中,抑制型和非抑制型基因频率分布在子痫前期组和正常晚孕组问差异均无统计学意义(P均〉0.05):母儿间KIR2D基因亚型与HLA—Cw等位基因特异性配伍分析中子痫前期组和正常晚孕组间差异均无统计学意义(P均〉0.05)。结论母儿间KIR2D和HLA—Cw基因多态性配对与子痫前期的发病无关。Objective To investigate the analyze association between killer cell immunoglobulin-like receptor(KIR)2D alleles combinated with human leucocyte antigen Cw (HLA-Cw) and preeclampsia in Chi- nese population. Methods The genetic typing was performed using PCR-sequence specific primer (PCR- SSP) technology in 117 preeclampsia pregnant women (preeelampsia group) and 106 normal pregnant women (normal pregnant group) with their babies. The genetic frequence and specific binding of KIR2D to HLA-Cw were analysed. Results The frequency of KIR2DS2 and HLA-Cwl2 in preeclampsia group (5.12% and 8.12% ) were significantly higher than in normal pregnant group ( 1.58% and 3.30% ) (Pall〈 0.05). In the analy- sis of KIR2D genotypes combination, the distribution of gene frequency in inhibitory type and non inhibitory type had no significant differences between preeelampsia group and normal pregnant group (Pall〉 0.05 ). And there were also no significant differences between maternal/fetus specific binding of KIR2D and HLA-Cw in two groups (Pall〉 0.05). Conclusion The polymorphism of KIR and HLA-Cw and their specific combination are not associated with the pathogenesis of preeclampsia.
