大鼠肝脏低温灌注和复流模型改进及B7表达的研究

摘  要:目的探讨共刺激分子B7—1和B7-2在大鼠肝脏低温灌注和复流模型中的表达及其免疫学意义。方法将30只大鼠随机分为3组:A组(低温灌注20min再复流24h)、B组(低温灌注30min再复流24h)、C组(假手术组)。分别取模型之肝脏,应用逆转录聚合酶链反应(PT-PCR)检测B7.1和B7-2在3组肝脏组织中mRNA表达。结果B7—1和B7-2mRNA在C组以极低水平表达(0.226±0.053,0.339±0.049),而在A组、B组表达却明显增多(P〈0.01),且B组明显高于A组(B7。1:0.475±0.079比0.3674-0.052,B7-2:0.666±0.039比0.595±0.050;P〈0.01)。结论低温灌注和复流之肝脏通过上调B7—1和B7-2的表达来增加肝脏的免疫原性。Objective To elucidate the expression of costimnlatory molecule B7-1 and B7-2 in hv- er hypothermic irrigation and reperfusion model of rats and its significance. Methods Thirty rats were ran- domly divided into three groups : group A, hypothermic irrigation for 20 min ; group B, hypothermic irriga- tion for 30 min; and group C, sham operation. The expression of B7-1 and B7-2 mRNA in the livers was detected by using real-time reverse transcription polymerase chain reaction (RT-PCR). Results The B7-1 and B7-2 expression levels were at very low ievels in the liver tissues of group C (0. 226 ~ O. 053, and 0. 339 _+0. 049), but the mRNA expression levels of B7-1 and B7-2 were significantly enhanced in group A or group B (P〈0. 01). In addition, both B7-1 and B7-2 levels were higher in group B than in group A (B7-1 : 0.475±0. 079 vs O. 367±0. 052; B7-2:0. 666 +0. 039 vs O. 595±0. 050;P 〈 0. 01 ). Conclu- sion Hypothermic irrigation and reperfusion may increase the immunogenticity of the liver by upregnlating the costimulatory molecules B7-1 and 137-2.
