摘 要:目的:建立同时测定冬虫夏草药材中腺苷、虫草素和麦角甾醇的方法。方法:Diamosil C18色谱柱(4.6 mm×250 mm,5μm),流动相甲醇-水梯度洗脱,体积流量1.0 mL.min-1,柱温30℃,进样量10μL,检测波长260 nm。结果:腺苷、虫草素和麦角甾醇分别在0.032 2~0.805,0.015 4~0.385,0.084 3~2.107 5μg线性关系良好,回归方程分别为Y=2 573X+0.737 8;Y=2 631X+0.624 4;Y=2 315X+0.482 9,平均加样回收率分别为98.94%(RSD 1.16%),99.73%(RSD 2.19%),99.55%(RSD 2.02%)。结论:该方法准确性高,重复性好,可作为冬虫夏草药材的质量控制方法。Objective: To establish the quantitative analysis method for adenosine, cordycepin and ergosterol in Cordyceps sinensis by HPLC. Method: HPLC was performed on Diamosil C18 column(4.6 mm×250 mm, 5 μm); the mobile phase consisted of methanol and water at a flow rate of 1.0 mL·min-1.The column temperature was set at 30 ℃, and the eluate was detected at 260 nm. Result: Quantitative analysis of HPLC showed that the linear ranges of adenosine, cordycepin and ergosterol were 0.032 2-0.805, 0.015 4-0.385 and 0.084 3-2.107 5 μg respectively.The average recoveries of adenosine, cordycepin and ergosterol were 98.94%(RSD 1.16%), 99.73%(RSD 2.19%), 99.55%(RSD 2.02%)respectively. Conclusion: The method set up is accurate, feasible and reliable, therefore available for the quality control of C. sinensis.
