摘 要:目的检测食管癌细胞株KYSE-150和正常食管上皮细胞株Het-1A中hsa-mir-1246的表达,并对hsa-mir-1246进行靶基因预测及相关生物信息学分析,为深入研究hsa-mir-1246在食管癌发生、发展过程中的分子机制提供理论依据和实验基础。方法应用qRT-PCR技术检测食管癌细胞株KYSE-150中hsa-mir-1246的表达水平,并与正常对照细胞株对比;通过miRBase、NCBI、UCSC Browser等在线工具分析hsa-mir-1246序列;应用TargetScan、miRanda及miRBD预测hsa-mir-1246的靶基因,进行功能富集分析和信号通路富集分析。结果 (1)食管癌细胞株中hsa-mir-1246表达水平明显高于正常对照细胞株(86.67±16.87 vs 30.95±11.51,P<0.05)。(2)miRNA-1246在人类同源的物种之间具有高度保守性。(3)3种预测网站得到hsa-mir-1246靶基因分别为114个、5 983个、398个,交集为13个。(4)hsa-mir-1246靶基因功能分别富集于转录的调控、信号转导、染色质修饰、细胞周期等生物学过程(P<0.001),涉及基底细胞癌、神经胶质瘤、Wnt信号通路、长时程增强效应等信号转导通路(P<0.05)。结论 hsa-mir-1246靶基因参与肿瘤发生的多种生物学过程,提示hsa-mir-1246可能与食管癌的发病机制密切相关。Objective To lay foundations and provide theoretical basis for the further studies on molecular mechanisms of hsa - mir - 1246 in human esophageal squamous carcinoma, this study investigated the expression of hsa - mir - 1246 in human esophageal squamous carcinoma cell line (KYSE - 150), and predicted its targets by bioinformatics analy- sis. Methods The expression of hsa - mir - 1246 was assessed by qRT - PCR. TargetSean, miranda and miRBD were used to predict target genes of hsa - mir - 1246, and the results was analyzed by Gene Ontology (GO) and pathway analysis. Results ( 1 ) The expression of hsa - mir - 1246 in human esophageal squamous carcinoma cell line was significantly higher than that in human esophageal epithelial cell line (86. 67± 16. 87 vs 30. 95 ± 11.51, P 〈0. 05). (2) The miR - 1246 was highly conserved among the species of the human homolog. (3) Thirteen target genes were predicted by the intersection of Targetsean, miRanda and miRBD. (4) Gene ontology analysis demonstrated that the target genes of hsa - mir - 1246 were enriched in regulation of transcription, signal transduetion, ehromatin modification, cell cycle and other biological processes (P 〈 0. 001 ). The pathway analysis indicated that the target genes mainly involved in basal cell carcinoma, glioma, Wnt signaling pathway, long - term potentiation and other pathways ( P 〈 0. 05 ). Conclusion The target genes of hsa - mir - 1246 are enriched in multiple biological processes relevant to tumor signaling pathways. Therefore hsa - mir - 1246 is probably involved in human esophageal squamous carcinoma pathogenesis.
