摘  要:目的观察博来霉素在体外对人皮肤恶性黑素瘤A875细胞增殖的影响,为临床化疗提供参考。方法体外培养A875细胞,分别给予不同浓度的博来霉素(0.5~32μg/ml)、不同时间(24 h或48 h)作用于细胞,倒置显微镜观察细胞形态,CCK-8法检测不同浓度的博来霉素作用不同时间对A875细胞增殖的影响。结果博来霉素使贴壁细胞密度下降,变圆、变形的漂浮细胞增多;博来霉素对A875细胞的生长增殖有明显的抑制作用,呈剂量和时间依赖性。结论博来霉素能有效抑制人皮肤恶性黑素瘤A875细胞的增殖。Objective To observe the effect of bleomycin(BLM) on the proliferation of human skin A875 malignant melanoma cells in vitro,and to provide reference for drug therapy in clinical practice. Methods A875 cells were cultured with different concentrations(0.5 -32 μg/ml) of BLM for 24 or 48 hours in vitro. The cellular morphology of A875 ceils were observed by light inverted microscope. The proliferation of A875 cells was examined by cell counting kit-8 (CCK-8) method. Results Decrease in adhesive cell density to the base of wells could be observed after being treated with BLM while increases in round and deformed floating cells could be observed. BLM had a strong effect of inhibition on A875 cells proliferation in a concentration-dependent and time-dependent manner. Condusion BLM can inhibit the proliferation of human skin A875 malignant melanoma cells significantly.
