摘  要:目的:检测IL-17在喉鳞状细胞癌(LSCC)及癌旁组织中的表达及在肿瘤相关性巨噬细胞(TAMs)上的分布特点。探讨在LSCC发生、发展中IL-17的表达与TAMs分布及新血管生成的关系。方法:采用SP技术对32例LSCC组织及20例癌旁组织进行免疫组织化学染色,检测IL-17、CD68及CD34的表达情况,测定微血管密度,分析LSCC组织黏膜炎症程度及各临床参数之间的关系,双重免疫组织化学染色用于观察IL-17在CD68+巨噬细胞的表达及定位。结果:①LSCC组织中IL-17、CD68、微血管密度、总炎症细胞数、腺体数较癌旁组明显增多(P<0.05)。②免疫组织化学结果显示IL-17在LSCC中高表达(P<0.01),在不同病理分化组中IL-17的表达差异有统计学意义(P<0.05),IL-17与CD68+细胞表达呈正相关(r=0.576,P<0.05)。③IL-17及CD68双重免疫组织化学染色证明IL-17主要定位于巨噬细胞。④IL-17的高表达与微血管密度正相关(r=0.743,P<0.05)。结论:IL-17在LSCC的发生、发展过程中可能通过影响肿瘤相关性巨噬细胞的功能及相关的细胞因子导致肿瘤炎症的发生。IL-17可能在LSCC的浸润转移中具有一定的作用。Objective:To detect the expression of interleukin 17(IL-17) in laryngeal squamous cell carcinoma (LSCC) and adjacent tissues of carcinoma. Correlation analysis the expression of IL-17 and the distribution of tumor associated macrophages in LSCC. Method: All the samples were came from China-Japan Union Hospital of Jilin University from March 2008 to September 2009,including 32 cases of squamous cell carcinoma of larynx and 20 cases adjacent tissues of carcinoma as control. Histopathological is used for routine tissue pathology classifica- tion and evaluated mucosal inflammation degree; Immunohistochemistry stain was used to determine IL-17, CD68, CD34 expression in 52 specimens. Double immunohistochemical staining was done to locate which cells expressed IL-17. Result: The inflammation response(totle imflamation cells) had increased in LSCC compare with control. Overexpression of IL-17 protein(P〈0.01) were detected in LSCC. The expression levels of IL-17 were different between well differentiation and mordenerat-poorly differentiation of LSCC group(P〈0.01 ). The IL-17 express- ing cells mainly were located in maerophages (CD68+/ILl7 +) as demonstrated by double immunohistochemical staining. IL-17 expression significantly correlated with MVD(CD34 + ) and TAMs(CD68 + ) in LSCC group( P〈0.05). Conclusion:With the development of laryngeal squamous cell carcinoma, the inflamation mediated by IL-17 may be realized through affecting the function of macrophage and some related cytokines. IL-17 play a certain role in the metastasis of squamous cell carcinoma of larynx.
