顺铂增加BCL-2蛋白高表达的鼻咽癌细胞凋亡

摘　　要：目的:通过对3种不同鼻咽癌细胞的BCL-2蛋白表达、细胞存活率和凋亡率的观察,探讨提高顺铂治疗鼻咽癌疗效的新方法。方法:采用Western blot技术检测3种不同鼻咽癌细胞CNE1、C666-1及SUNE1的BCL-2蛋白的表达情况;采用MTT法及流式细胞技术,检测并比较顺铂对3种鼻咽癌细胞的细胞存活率和凋亡率的影响。结果:BCL-2蛋白在CNE1及C666-1细胞上高表达,在SUNE1上几乎无表达(P<0.05);经顺铂干预后,CNE1及C666-1细胞的细胞凋亡率明显升高,存活率明显低于对照组(P<0.05);SUNE1细胞的细胞凋亡作用较弱(P<0.05)。结论:BCL-2蛋白在不同类型的鼻咽癌细胞上表达量不同,提示顺铂对BCL-2蛋白高表达的鼻咽癌细胞的杀伤作用更强。Objective：To explore a new target of cisplatin-Induced apoptosis. Method：The expressions of BCL- 2 protein in three types of nasopharyngeal carcinoma cells CNE1, C666-1 and SUNE1 were detected by Western blot; after treated with cisplatin, the survival rates of the three cells were evaluated with MTT assay, and the ap- optosis rates were detected by flow cytometr. Result：BCL-2 protein was highly expressing in CNEI,C666-1, while there was nearly no in SUNE1 （P〈0.05）. The survival rates of CNE1 and C666-1 were significantly increased and the apoptosis rates were decreased obviously after treatea with cisplatin（P〈0.05）, while the effect of the cis- platin was poor on SUNE1. Conclusion： The expression of BCL-2 protein was different in different type of NPC cells; the NPC BCL-2 protein highly experssion were sensitived to cisplatin-induced apoptosis.
