海水干预下人肺泡上皮细胞低氧诱导因子1α与TNF-α、IL-6表达的变化

摘　　要：目的:探讨海水干预对人肺泡上皮细胞低氧诱导因子1α蛋白及TNF-α、IL-6表达的影响。方法:肺泡上皮细胞来源的A549细胞系,分为正常对照组(C)和海水处理组(S)。C组用新鲜培养基常规培养,S组经灭菌配方海水孵育0.5、1、2、4、8、16h,蛋白免疫印记法(western blot)检测各组细胞HIF-1α蛋白的表达,放射免疫法检测细胞培养上清液中TNF-α、IL-6的含量。结果:(1)HIF-1α在海水干预1h后开始升高,4h达高峰(P<0.01),此后下降,但仍显著高于对照组(P<0.01)。(2)TNF-α与IL-6在海水干预1h后开始升高,2h达高峰(P<0.01),4h及8h组较对照组无统计学差异。结论:海水干预可诱导肺泡上皮细胞HIF-1α蛋白及炎性因子TNF-α、IL-6的表达;HIF-1α在海水干预致肺泡上皮细胞炎症反应过程中可能起重要作用。Objective To explore the effects of seawater on the expressions of hypoxia-indueible factor-1α, TNF-α and IL-6 in human alveolar epithelial ceils （A549）. Methods The A549 cells were divided into two groups： control group （C） and seawater treatment group （S）. The cells of the seawater treatment group were cultured in sterile dispensed seawater, while the cells of the control group were cultured in DMEM. The expressions of HIF-1α in the two groups were detected by western blot analysis. The levels of TNF-α and IL-6 in the cell culture supernatants in the two groups were measured by radioimmunoassay. Results The expression of HIF-1α in seawater treatment group increased after 1 h, reached the peak at 4 h （P 〈 0.01 ）, then decreased, but still higher than that in control group （P 〈 0.01 ）. The expressions of TNF-α and IL-6 in seawater treatment group increased after 1 h, reached the peak at 2 h （P 〈 0.01 ）, then decreased. After 4h or 8h culture, the expressions of TNF-α and IL-6 didn＇t show statistical differences between the two group. Conclusions Seawater treatment can induce the expressions of HIF-1α, TNF-α and IL-6 in alveolar epithelial cells. HIF-1α may play an important role in the inflammatory response in alveolar epithelial cells induced by seawater.
