摘 要:目的:探讨生殖细胞核因子(GCNF)在宫颈癌中的表达及其在宫颈癌发生发展的作用。方法:运用免疫组织化学Envision法和RT-PCR技术检测GCNF蛋白和mRNA在正常宫颈组织、上皮内瘤变组织及宫颈鳞癌组织中的表达。结果:GCNF蛋白在正常宫颈组织、上皮内瘤变组织(CINⅠ、CINⅡ、CINⅢ)和宫颈鳞癌组织中的表达率分别为90.0%(18/20)、78.3%(18/23)、45.0%(9/20)、33.3%(9/27)和3.4%(4/29),差异有统计学意义(P<0.05)。RT-PCR结果显示GC-NF mRNA的表达与GCNF蛋白相一致,即宫颈鳞癌组织与正常宫颈组织相比GCNF mRNA的表达显著降低(P<0.05)。结论:初步认为GCNF蛋白和mRNA表达的下调或缺失可能是宫颈癌发生的一个早期信号,以期为宫颈癌的靶向治疗提供研究方向。Objective: To explore the expression of germ cell nuclear factor (GCNF) in cervical cancer and its role in oncogenesis and development of cervical cancer. Methods: The expressions of GCNF protein and GCNF mRNA were detected by imnmnohistochemical Envision method and RT- PCR technique in normal cervical tissue, cervical intraepithelial neoplasia (CIN) tissue (CIN I , CIN II , CIN III ), and cervical cancer tissue. Results: The expression of GCNF protein in normal cervical tissue, CIN and cervical cancer were 90. 0% (18/20), 78.3% ( 18/23), 45.0% (9/20), 33.3% (9/27), and 3.4% (4/29), respectively, there was statistically significant differ- ence ( P 〈 0. 05 ) . The results of RT - PCR showed the expression of GCNF mRNA was consistent with GCNF protein, tile expression of GCNF mRNA in cervical cancer was statistically significantly lower than that in normal cervical tissue. Conclusion : PreliminaJy view is that the down - regulation or deletion of expressions of GCNF protein and mRNA may be an early signal of cervical cancer. It may provide research direction for targeted therapy of cervical cancer.
