摘 要:目的探讨生物钟基因clock在脑胶质瘤发生、发展中的作用。方法采用RT-PCR及免疫组化方法检测67例不同级别的脑胶质瘤(胶质瘤组)和周围正常组织(对照组)clock的转录活性及表达强度。结果两组clock基因均有表达,胶质瘤组Ⅲ~Ⅳ级者clock基因mRNA及蛋白表达均高于Ⅰ~Ⅱ级者及对照组(P均<0.05)。结论生物钟基因clock在高级别脑胶质瘤组织中呈高表达,clock表达强度与脑胶质瘤的恶性程度呈正相关。Objective To study the significance of human circadian clock genes in the development and progression of gliomas. Methods Immunohistoehemieal was conducted to detect the transcriptional activity and expression intensity of clock protein of gliomas ( gliomas group) and adjacent tissues( control group) in 67 gliomas patients. RT-PCR was conduc- ted to detect clock gene mRNA. Results The expression of clock was positive detected in both two groups. Clock mRNA and protein were both higher in grade Ⅲ and IV glioma group than those of grade I and 1I as well as contorl group ( all P 〈 0.05 ). Conclusions There was higher expression of clock in high grade gliomas than that of normal brain tissues. The expression intensity of clock genes may be positively related to the malignancy of gliomas.
