摘 要:目的分析血清α1抗胰蛋白酶(α1-AT)和α1酸性糖蛋白(α1-AGP)在儿童肾母细胞瘤血清中的表达,并探讨筛查的临床价值。方法选取2007年1月至2010年12月就诊的38例肾母细胞瘤儿童为观察组,38例健康儿童为对照组。使用荧光定量PCR分析两组儿童血清中α1-AT和α1-AGPmRNA转录水平,使用ELISA法检测血清中α1-AT和α1-AGP的浓度。结果观察组血清中α1-AT和α1-AGPmRNA相对量分别为3.50±0.37、2.10±0.41,均显著高于对照组的0.90±0.45、0.50±0.24,差异有统计学意义(P=0.000、0.002)。观察组血清中α1-AT和α1-AGP浓度分别为(4516.8±102.4)、(1316.0±27.3)g/L,均显著高于对照组的(2467.4±23.8)、(728.6±9.4)g/L,差异有统计学意义(P=0.015、0.008)。结论血清α1-AT和α1-AGP检测可用于儿童肾母细胞瘤的诊断,且可应用于儿童。肾母细胞瘤的早期筛查。Objective To analyze the serum α1-antitrypsin (α1-AT) and α1- glycoprotein ( α1-AGP ) in children with Wilms tumor, and to explore the clinical application value. Methods Thirty- eight children with Wilms tumor who received treatment from January 2007 to December 2010 were designed as observation group, 38 healthy children were designed as control group. Fluorescence quantitative PCR was performed to analyze α1-AT and α1-AGP mRNA transcription levels of two groups, and ELISA was used to measure the serum concentration of α1-AT and α1-AGP. Results The mRNA levels of α1-AT and α1-AGP in observation group were 3.50 ± 0.37 and 2.10 ± 0.41 ,which were significantly higher than those in control group(0.90 ± 0.45,0.50 ± 0.24) (P= 0.000,0.002). The levels of α1-AT and α1-AGP in observation group were (4516.8 ± 102.4), (1316.0 ± 27.3) g/L, which were significantly higher than those in control group [ ( 2467.4 ± 23.8 ), (728.6 ± 9.4) g/L ] (P = 0.015,0.008 ). Conclusion To detect the levels of α1-AT and α1-AGP have the potential value for the diagnosis of Wilms tumor, which can be applied to early screening.
