摘 要:目的:观察人牙髓干细胞(human dental pulp stem cells,hDPSCs)表面是否有Ⅰ型趋化因子受体(CXCR1)表达、人工重组白细胞介素8(IL-8)对hDPSCs迁移和增殖的影响。方法:采用间接免疫荧光技术检测hDPSCs上CXCR1的表达情况;采用CCK8、体外趋化实验观察不同浓度人工重组IL-8对hDPSCs迁移和增殖的影响。结果:在正常hDPSCs细胞膜表面有CXCR1的表达。200、500 ng/mL IL-8作用4 h可以显著趋化hDPSCs的迁移(P<0.05)。10、50、100 ng/mL IL-8作用48 h可促进hDPSCs增值(P<0.05)。结论:CXCR1在hDPSCs细胞膜上表达,IL-8能够促进hDPSCs的迁移和增殖。AIM: To observe the expression of CXCR1 in human dental pulp stem cells (hDPSCs) in vitro and explore the role of IL-8 on the migration and proliferation of hDPSCs. METHODS : CXCR1 expression in hDPSCs was detected by indirect immunofluorescence technique. The effects of recombinant human IL-8 on the migration and proliferation of hDPSCs at different concentrations were observed by transwell chamber assay and CCK8 Assay. RE-SULTS: CXCR1 was expressed on the cytomembrane of hDPCSs. IL-8 at 200ng/mL and 500 ng/mL increased the chemotactic migration of hDPSCs after 4 h incubation (P 〈 0.05). After incubation with IL-8 at 10 ng/mL, 50 ng/mL and 100 ng/mL for 48 hs the proliferation of hDPSCs was increased (P 〈0.05), CONCLUSION: hDPSCs express CXCR1. IL-8 can accelerate the migration and the proliferation of hDPSCs.
