摘  要:目的:通过NF-κB(nuclear factor-κB,NF-κB)抑制剂(Pyrrolidine dithiocarbamate,PDTC)研究NF-κB信号通路在人牙髓细胞分化中的作用。方法:组织块酶消化法分离培养人牙髓细胞,并用含有20μmol/L PDTC的矿化液对其进行诱导培养2周,RT-PCR检测PDTC刺激对牙髓细胞矿化相关基因碱性磷酸酶(Alkaline phosphatase,ALP)、骨钙素(Osteonectin,OCN)和骨桥素(Osteopontin,OPN)mRNA表达水平的影响;诱导培养4周后茜素红染色观察PDTC刺激下牙髓细胞形成矿化结节情况。结果:矿化诱导培养2周后,RT-PCR检测显示:PDTC刺激组的ALP、OCN和OPN mRNA的表达水平均明显上调,分别与对照组相比,差异均有统计意义(P<0.05)。矿化诱导培养4周后,茜素红染色显示:PDTC刺激组矿化结节明显多于对照组。结论:PDTC可促进牙髓细胞分化,提示NF-κB信号通路可能具有抑制牙髓细胞分化的作用。AIM: To investigate the effect of nuclear factor-κB ( NF -κB) signaling pathway on the differ-entiation of human dental pulp cells (hDPCs). METHODS: Tissue block enzymolytic method was used to isolate and culture hDPCs, hDPCs of passage 4 were stimulated with inducing culture medium containing 20 μmol/L of pyrolidine dithiocarbamate (PDTC), an NF-κB inhibitor. Cells cultured with no PDTC served as the controls. 2 weeks after min- eralization induction culture, mRNA expressions of osteonectin (OCN), alkaline phosphatase (ALP) and osteopontin (OPN) were detected by RT - PCR. 4 weeks after induction mineral nodules were observed by alizarin red S staining. RESULTS: OCN, ALP and OPN mRNA expressions were significantly increased in the PDTC treated cells ( P 〈 0.05 ). More mineral nodules were observed in PDTC treated cells than in the controls ( P 〈 0.05 ). CONCLU-SION: NF-κB signaling pathway might play an inhibitory role on the differentiation of human dental pulp cells.
