摘  要:目的:研究增殖细胞核抗原(PCNA)及人类N-myc下游调节基因1(NDRG1)在人肝细胞性肝癌中的表达情况,探讨其与肝癌生物学行为的关系及临床意义。方法:选择有存档的原发性肝癌标本58例,肝硬化34例,正常肝组织标本15例,用H-E染色观察组织形态,用免疫组织化学SABC检测PCNA和NDRG1的表达。结果:肝癌组织中PCNA表达明显高于肝硬化组织和正常组织;在肝硬化组织和正常肝组织中,PCNA的表达没有差异;肝癌中PCNA的表达与患者的性别、年龄、HbsAg阳性、AFP水平、部位和肿瘤的直径无关。NDRG1在正常肝组织、肝硬化组织、肝癌组织中表达逐渐减弱;肝硬化组与正常肝组织组相比,差异无统计学意义,在肝癌组织中NDRG1与患者的性别、年龄、HbsAg阳性、AFP水平、部位和肿瘤的直径无关。PCNA及NDRG1在肝癌组织中的表达呈负相关。结论:PCNA、 NDRG1在肝癌发生、发展过程中起着重要的作用,联合检测可以为肿瘤的早发现、早诊断、早治疗提供判断依据。Objective: To study the expression of proliferating cell nuclear antigen (PCNA) and N-myc downstream regulated gene 1 (NDRG1) in human primary hepatocellular carcinoma, and to confer the biological behavior and the clinical significance in the hepatocellular carcinoma Methods: Documented specimens were selected, containing 58 cases of primary liver cancer, 34 cases of cirrhosis and 15 eases of normal tissues. H-E staining was performed to observe the morphology, and irnmunohistochemistry SABC method to study the expression of tK;NA and NDRG1 in primary liver cancer. Results: The expression rate of PCNA in primary liver cancer was significantly higher than that of liver cirrhosis tissues and normal liver tissues. The expression of PCNA in hepatocellular carcinoma was not related to the patient gender, age, HbsAg-positive, AFP level, independent of location and diameter of the tumor. NDRG1 expression was gradually decreased in the normal liver tissues, cirrhesis liver tissues and primary liver tissues; There were no signifieant difference between the cirrhosis group and the normal liver tissue group; The expression of NDRG1 in hepatocellu- lar carcimoma was not related to the patient gender, age, HbsAg-positive, AFP level, in dependent of location and diameter of the tumor. The expression of NDRG1 in HCC was negatively correlated with PCNA in this experiment. Conclusion : PCNA and NDRG1 in hepatocellular carcinoma play an important role in their development processes. Joint detection contributes to the early cancer detection, early diagnosis and early treatment.
