三物白散含药血清对胃癌MGC-803细胞增殖、凋亡及survivin基因表达的影响

摘　　要：目的研究三物白散含药血清对人胃癌MGC-803细胞株的增殖、凋亡及survivin基因表达的影响。方法体外实验选择MGC-803细胞,以5-氟尿嘧啶为阳性对照,采用MTT比色及吖啶橙、EB染色法检测低、中、高3组三物白散大鼠含药血清对MGC-803细胞增殖、凋亡的影响,运用RT-PCR法检测survivinmRNA的表达。结果三物白散含药血清作用MGC-803细胞后,随着三物白散含药血清药物浓度的增加及作用时间的延长,细胞增殖率逐渐降低,细胞抑制率、凋亡率逐渐升高,survivin蛋白的表达相应的降低。结论三物白散含药血清能够抑制人胃癌MGC-803细胞增殖,对细胞有诱导凋亡作用,在凋亡过程中涉及sur-vivin基因表达下降。Objective： To tudy the efection of Sanwubai Powder medicated serum on proliferation,apoptosis and survivin expression of gastric cancer cell Lines MGC-803.Methods： MGC-803 in vitro was selected.It was used the 5-Fu as the positive control.The proliferation and the apoptosis of the Sanwubai Powder containing serum on the MGC-803 with the low,the medium and the high dose were detected by the colorimetric MTT and acridine orange,EB staining methods.And mRNA expression of survivin were detected by RT-PCR method.Results： After Sanwubai Powder medicated serum treatment to Gastric cancer cell MGC-803,as Sanwubai Powder drug-containing serum drug concentration increasing and time extending,cell proliferation rates decreased,cell inhibition rate and the rate of apoptosis increased gradually,the expression of survivin protein was lower correspondly.Conclusion： Sanwubai Powder drug-containing serum can inhibite proliferation of human gastric cancer MGC-803 cell,promote cell apoptosis.The expression of survivin gene will reduce in the apoptotic process.
