摘 要:【目的】研究胃癌细胞-树突状细胞(Dc)融合疫苗对严重联合免疫缺陷(scID)小鼠胃癌模型的抗肿瘤效应。【方法】聚乙二醇法诱导胃癌细胞BGC-823与成熟DC融合制备疫苗,将SCID小鼠胃癌模型随机均分为4组:BGC-823/DC融合疫苗组、BGC-823+DC联合组、DC组和PBS组,分别皮下注射相应疫苗、细胞或试剂,检测小鼠脾脏细胞毒性T淋巴细胞(CTL)活性、皮下移植瘤体积、核因子-KB(NF-KB)、基质金属蛋白酶-9(MMP-9)表达和癌细胞侵袭能力即穿膜细胞数。【结果】CTL活性在融合疫苗组、联合组、DC组及PBS组分别为(39.40±3.38)%、(18.07±1.49)%、(14.074±1.32)Vo、(10.70±1.11)%;移植瘤体积在以上四组分别为(165.03±30.96)mm3、(767.65±118.81)mm3、(1025.16±80.90)mnl。、(1314.99士74.03)mm3;NF-KB表达分别为20.0%、70.0%、90.0%、100.0%;MMP-9表达分别为50.0%、80.0%、90.0%、90.0%;穿膜细胞数分别为15.17±4.67,39.73±6.09、59.36±13.87、90.86±4.97。融合疫苗组各检测指标与其他组比较均有显著差异(P〈O.05)。【结论】融合疫苗能有效诱导CTL活化、下调NF—KB、MMP-9表达及癌细胞侵袭能力,产生特异性抗肿瘤免疫。[Objective] To investigate anti-tumor effect of gastric carcinoma cell-dendritic cell (IX]) fusing vac- cine on severe combined immunodeficiency(SCID) in mouse gastric carcinoma model. [Methods] Vaccine was achieved by the fusion of BGC-823 gastric carcinoma cell with maturate DC by using polyethyleneglycol method. SCID mouse gastric carcinoma models were divided randomly and equally into four groups including BGC-823/DC fusing vaccine group, BGC-823+DC combining group, DC group and PBS group, and then injected subcutaneously with the corre- sponding vaccine, cell or reagent. Cytotoxic T cell(CTL) activity in mouse spleen, volume of subcutaneous transplan- tation tumor, expressions of nuclear factor-~B (NF-~B) and matrix metalloproteinase-9 (MMP-9) and cancer cell inva- sion (i. e. number of countersinking-membrane cells) were detected. [Results] CTL activity in fusing vaccine group, combining group, DC group and PBS group were (39. 404-3. 38)o//00, (18. 074± 1.49)%, (14.07± 1.32)% and (10.70 ±1. 11)%, respectively. Volumes of subcutaneous transplantation tumor in four groups above-mentioned were (165. 034-30.96)mm3, (767.65±118.81)mm3, (1025. 164±80.90)mm3 and (1314. 994±74.03)mm3, re- spectively. Expressions of NF-B were 20.0%, 70.0%, 90.0% and 100.0%, respectively, and those of MMP-9 were 50.0%, 80.0%, 90.0% and 90.0%, respectively. Numbers of countersinking-membrane cells were 15.17:k 4.67, 39.73 ±6.09, 59.36 4±13.87 and 90.86 ± 4.97, respectively. There was Significant difference in the detection indexes between fusing vaccine group and other groups( P d0.05). [Conclusion] Fusing vaccine can effectively in- duce the activation of CTL, and down-regulate the expressions of NF-B and MMP-9 and malignant cell invasion, and elicit specific anti-tumor immunity.
