胃腺癌组织中SLP-2的表达及其对SGC7901细胞增殖能力的影响

摘　　要：目的:探讨胃腺癌组织中SLP-2蛋白的表达情况及沉默SLP-2对胃腺癌SGC7901细胞增殖的影响。方法:采用免疫组化法检测45例胃腺癌患者的癌组织、配对癌旁正常胃黏膜组织中SLP-2蛋白的表达情况,分析其与临床病理特征的关系;将SLP-2siRNA转染SGC7901细胞,采用MTT法和流式细胞术检测转染后细胞的增殖状况及细胞周期。结果:SLP-2蛋白在胃腺癌组织中的阳性表达率(68.9%,31/45)高于癌旁正常组织(26.7%,12/45)(χ2=15.429,P<0.001)。胃腺癌组织中SLP-2的表达与淋巴结转移及TNM分期密切相关(校正χ2=4.784,5.318,P均<0.05)。SLP-2siRNA转染SGC7901细胞后,细胞增殖活性降低;G1细胞期增多,S期细胞减少(F=14.459,11.825,P均<0.05)。结论:SLP-2参与了胃腺癌的发生、发展。Aim：To investigate the expression of SLP-2 in gastric adenocarcinoma and its effects on proliferation of SGC7901 cell line.Methods：The expression of SLP-2 in 45 cases of gastric adenocarcinoma tissue and adjacent tumor-free tissue was detected by immunohistochemistry,and its association with clinical pathological characteristics was also analyzed.SLP-2 siRNA was designed and transfected into gastric adenocarcinoma cell line SGC7901.The cell proliferation was detected by MTT assay,and the cell cycles was analyzed by flow cytometry.Results：The positive rate of SLP-2 expression in gastric adenocarcinoma was 68.9%（31/45）,which was significantly higher than that of the adjacent tumor-free tissue（26.7%,12/45）（χ2=15.429,P0.001）.The expression of SLP-2 in gastric adenocarcinoma tissue was associated with lymph nodes metastasis and TNM staging（χ2=4.784,5.318;both P0.05）.After SLP-2 siRNA was transfected,the number of cells in G1 phase was increased and S phase was declined（F=14.459,11.825;both P0.05）.Conclusion：SLP-2 protein shows up-regulation in gastric adenocarcinoma,and it may have some relationship with carcinogenesis and development of gastric adenocarcinoma.
